Intramolecular Palladium-Catalyzed Carboamination for the Stereoselective Synthesis of Five-Membered Iminosugar C-Glycosides

Summary: In this study, we present a novel strategy for the stereoselective synthesis of five-membered iminosugar C-glycosides using an intramolecular palladium-catalyzed carboamination reaction. We successfully synthesized two sugar-derived aminoalkenes, which were then subjected to carboamination conditions with various aryl bromides. This resulted in the formation of a library of protected iminosugars carrying a 1-C-arylmethyl substituent, some of which were fully deprotected to yield original iminosugar C-glycosides. This methodology offers a highly efficient and concise pathway to synthesize these important molecules.

Biocatalytic Cascades toward Iminosugar Scaffolds Reveal Promiscuous Activity of Shikimate Dehydrogenases

Summary: In this study, a protecting group-free chemoenzymatic and biocatalytic cascade was developed to synthesize iminosugars from sugar-derived aminopolyols in two steps. The use of galactose oxidase variant F-2 followed by a chemical or enzymatic reduction allowed for efficient one-pot synthesis of these targets, with a product formation rate of over 70%. Genome mining was utilized to identify bacterial shikimate dehydrogenases as promiscuous iminosugar reductases, which was crucial for the success of this strategy. The cell-free protocols enabled isolation of highly polar iminosugar products from biotransformations in a single step through the development of a gradient-elution cation exchange purification. This two-step pathway provides a short synthetic route that can serve as a cell-free platform for broader iminosugar synthesis.

Morphological Plasticity of LiCl Clusters Interacting with Grignard Reagent in Tetrahydrofuran

LiCl aggregates as compact cubane-like Li4Cl4 structures, with an open-edge pseudotetrahedral frame promoted by solvent-assisted Li-Cl bond cleavage. LiCl prefers to coordinate MgCl2 through μ(2)-Cl bridges. The study identifies the role of LiCl in the determination of compounds present in turbo Grignard solutions.

Flow chemistry based catalytic hydrogenation for improving the synthesis of 1-deoxynojirimycin (DNJ) from an L-sorbose derived precursor

Stereoselective Access to Iminosugar C,C-Glycosides from 6-Azidoketopyranoses

Forodesine and Riboprine Exhibit Strong Anti-SARS-CoV-2 Repurposing Potential: In Silico and In Vitro Studies

Summary: Recently, nucleos(t)ide antivirals have emerged as top options for treating COVID-19 caused by SARS-CoV-2 infection. This study investigated various nucleoside analogues and identified riboprine and forodesine as highly effective in inhibiting the replication of the Omicron strain of SARS-CoV-2. These compounds showed superior activity compared to remdesivir and molnupiravir, and their active metabolites also displayed promising results. The findings suggest the repurposing potentials of riboprine, forodesine, and their nucleotides for the treatment of COVID-19.
