Asymmetric Synthesis of ?-Spiro-?-lactones and ?-Substituted ?-Lactones via Chiral Bifunctional Sulfide-Catalyzed Bromolactonizations

Summary: An efficient enantioselective synthesis of important pharmaceutical building blocks called gamma-chiral alpha-spiro-gamma-lactones was achieved through a catalytic bromo-lactonization reaction using BINOL-derived chiral bifunctional sulfide. This reaction is suitable for alpha-allyl carboxylic acids containing different structures. The resulting alpha-spiro-type bromolactonization product can be transformed to obtain optically active gamma-functionalized alpha-spiro-gamma-lactones. This catalytic system was also proven useful in the asymmetric synthesis of alpha,alpha-diaryl- and dialkyl-substituted gamma-lactones.

Summary: Carbon dioxide (CO2) is commonly used as a sustainable C1 synthon for synthesizing carboxylic acid derivatives, but its high stability and inertness limit its application in organic synthesis. Recent advances have successfully reduced the inert CO2 molecule using various approaches, including thermochemical, photochemical, and electrochemical methods. These greener approaches have revolutionized the synthesis of carboxylic acids under mild reaction conditions by incorporating CO2 into organic compounds via functionalization of (sp3)C-H bonds.

Differentiating the Allyl and Propargyl Groups on α-Quaternary Carboxylic Acids via Chiral Bifunctional Sulfide-Catalyzed Kinetic Resolution

Summary: Catalytic kinetic resolution is a reliable method for enantioselective synthesis of chiral molecules. However, it is challenging to perform kinetic resolution on racemic compounds with similar substituents at a chiral center. In this study, a kinetic resolution of a-allyl-a-propargyl carboxylic acids was achieved using a chiral bifunctional sulfide catalyst. The utility of the resulting optically active compounds was also demonstrated.

Bifunctional Onium and Potassium Iodides as Nucleophilic Catalysts for the Solvent-Free Syntheses of Carbonates, Thiocarbonates, and Oxazolidinones from Epoxides

Summary: The catalytic potential of organo-onium iodides in the coupling reactions of epoxides and carbon dioxide (CO2) was demonstrated. However, harsh reaction conditions were required for efficiently promoting these reactions. To overcome this issue, bifunctional onium iodide nucleophilic catalysts were developed. The successful design of these catalysts enabled nucleophilic catalysis under mild conditions, leading to the synthesis of 2-oxazolidinones and cyclic thiocarbonates from epoxides.

Efficient methods for the synthesis of chiral 2-oxazolidinones as pharmaceutical building blocks

Summary: Chiral 2-oxazolidinones are considered to be important heterocycles in medicinal chemistry, and many important pharmaceutical molecules are based on them. Therefore, the development of more efficient catalytic methods for synthesizing chiral 2-oxazolidinones is crucial in synthetic organic chemistry. This review summarizes the coupling reactions of epoxides and isocyanates for the preparation of 2-oxazolidinones, and introduces a synthetic example of a commercially available pharmaceutical compound using this method.

Enantioselective iodolactonization to prepare ε-lactone rings using hypervalent iodine

Summary: In recent years, enantioselective halolactonization reactions have grown rapidly, but most are only effective in forming smaller rings (6,5,4-membered). The formation of highly selective seven-membered epsilon-lactones is rare and has never been achieved without conformational bias. This study describes the first highly enantioselective 7-exo-trig iodolactonizations of conformationally unbiased epsilon-unsaturated carboxylic acids, using a combination of a bifunctional BAM catalyst, I-2, and I(iii) reagent (PhI(OAc)(2):PIDA).
