Evaluation of N, O-Benzamide difluoroboron derivatives as near-infrared fluorescent probes to detect β-amyloid and tau tangles

Yimin Chen et al.

Summary: This study introduced new N, O-coordinated organo-difluoroboron probes as binding scaffolds for Aβ plaques and Tau tangles, exhibiting suitable optical properties for NIR imaging. The probe 4PmNO-2 demonstrated excellent fluorescence capabilities and high affinity for AD biomarkers, making it a potential useful tool for in vivo NIR fluorescent imaging studies.

Near-Infrared Fluorescent Probes as Imaging and Theranostic Modalities for Amyloid-Beta and Tau Aggregates in Alzheimer's Disease

Himanshu Rai et al.

Summary: This paper discusses the limitations of current diagnostic tools for Alzheimer's disease and proposes the potential of small-molecular near-infrared probes as inexpensive and portable imaging tools. It summarizes the biochemistry of amyloid-beta and tau proteins and provides information on the spectroscopic properties and therapeutic effectiveness of NIR probes, aiming to guide the design of novel in vivo imaging fluoroprobes with theranostic capabilities in the future.

Oxidative Stress in Tauopathies: From Cause to Therapy

Fernando Bartolome et al.

Summary: Oxidative stress is the imbalance between reactive oxygen species (ROS) production and the antioxidant capacity of cells. The human brain is highly susceptible to oxidative stress, making it an essential component in the pathophysiology of neurodegenerative diseases such as tauopathies. However, there currently exist no effective therapies for these diseases. Although antioxidant therapies have shown promise in preclinical studies, clinical trials have been disappointing. To develop effective antioxidant therapies, a complete understanding of the molecular mechanisms underlying oxidative stress in tauopathies is crucial.

Alzheimer's disease and its treatment by different approaches: A review

Sukriti Srivastava et al.

Summary: Alzheimer's disease is a neurodegenerative disorder that impairs mental ability development and interrupts neurocognitive function. Current treatments only provide symptomatic relief with no cure available yet. Nanotechnology and stem cell-based therapy are seen as promising approaches to overcome the obstacles in treating AD.

Summary: The selenium-containing compound 13f, based on ebselen and verubecestat, showed potential anti-Alzheimer's disease and antioxidant stress effects by reducing beta-amyloid production and alleviating oxidative stress. Furthermore, 13f also demonstrated anti-inflammatory and cytoprotective effects, making it a promising candidate for AD treatment.

Linking Oxidative Stress and Proteinopathy in Alzheimer's Disease

Chanchal Sharma et al.

Summary: Proteinopathy and excessive production of reactive oxygen species are key factors contributing to neuronal toxicity in Alzheimer's disease. Beta-amyloid and tau proteins play a significant role in proteinopathy in AD, which is closely related to ROS production. The mechanisms of proteinopathy and ROS production work together to drive the pathology observed in aging brains, ultimately leading to oxidative stress and neurodegeneration.
