Summary: The ligand binding domain (LBD) of RORγt has been explored as a promising target for treating autoimmune diseases by modulating the production of IL-17 and IL-22 by Th17 cells. Various chemical scaffolds of small molecules have been discovered by multiple groups using methods such as HTS and virtual screening. Some of these compounds have entered clinical trials, showing potential for developing new RORγt modulators with higher potency and reduced toxicity.

Orthosteric and Allosteric Dual Targeting of the Nuclear Receptor RORγt with a Bitopic Ligand

Summary: The ROR gamma t nuclear receptor plays a critical role in Th17 cell differentiation and proliferation, as well as the production of the pro-inflammatory cytokine IL-17a. Small molecule inhibition of ROR gamma t is a promising strategy for treating autoimmune diseases. Developing bitopic ligands for ROR gamma t, capable of engaging both binding pockets, may provide advantages over traditional monovalent targeting strategies.

Cooperativity between the orthosteric and allosteric ligand binding sites of RORγt

Summary: Cooperative ligand binding is an important phenomenon in biological systems, where the mechanism behind it remains poorly understood. Through studying ROR gamma t and solving 12 crystal structures bound to various orthosteric and allosteric ligands, it was found that the orthosteric ligand induces a clamping motion of the allosteric pocket via helices 4 to 5, with Ala355 shifting between helix 4 and 5.
