Mercury Chloride Affects Band 3 Protein-Mediated Anionic Transport in Red Blood Cells: Role of Oxidative Stress and Protective Effect of Olive Oil Polyphenols

Summary: Mercury exposure induces oxidative stress in red blood cells (RBCs), but natural antioxidants like hydroxytyrosol (HT) and homovanillyl alcohol (HVA) from extra-virgin olive oil (EVOO) have shown potential protective effects. In this study, RBCs treated with HgCl2 were examined, and it was found that HT and/or HVA pre-treatment impaired acanthocytes formation and oxidative stress injury induced by mercury, while restoring the endogenous antioxidant system. These findings suggest that dietary supplementation with polyphenols could be beneficial for populations exposed to mercury poisoning.

Production of hydroxytyrosol rich extract from Olea europaea leaf with enhanced biological activity using immobilized enzyme reactors

Summary: This study developed and applied immobilized enzyme batch bioreactors for the chemo-enzymatic treatment of aqueous Olea europaea leaf extract. The modified extract showed increased enrichment in hydroxytyrosol and higher antioxidant, antibacterial, and cytotoxic activities compared to the non-modified extract. The biocatalyst demonstrated high hydrolytic stability and achieved high conversion yields of oleuropein during the modification process.

Single-Cell-Based Screening and Engineering of D-Amino Acid Amidohydrolases Using Artificial Amidophenol Substrates and Microbial Biosensors

Summary: Enantiomerically pure D-amino acids are crucial intermediates for synthesizing peptidomimetics and semisynthetic antibiotics. In this study, a transcriptional factor-based strategy was used to screen D-stereospecific amino acid amidase, which can be indicated by an enzyme-specific amidophenol substrate. Rapid directed evolution of the enzyme was achieved through biosensor cells, leading to the identification of a mutant with significantly improved catalytic activity towards various D-amino acids.

Improving bioconversion of eugenol to coniferyl alcohol by constitutive promoters in Escherichia coli

Summary: Coniferyl alcohol, a common monolignol precursor of lignin, is rarely found as free molecules in plants due to its conversion by peroxidases or laccases. In this study, 20 promoters were evaluated for expressing genes in Penicillium simplicissimum encoding vanillyl alcohol oxidase (PsVAO) and catalase (ScCTA1) to produce coniferyl alcohol while removing harmful H2O2. Through the construction of 100 combinations of constitutive promoters in E. coli, gene expression was enhanced, and coniferyl alcohol titers, bioconversion rates, and productivity levels were improved to 53.90 g/L, 86.72%, and 2.45 g/(L x h) respectively, using orthogonal experiments, fed-batch kinetic models, and the CRISPR/Cas9 system.
