Early-life low-level lead exposure alters anxiety-like behavior, voluntary alcohol consumption and AC5 protein content in adult male and female C57BL/6 J mice

Summary: Despite efforts to eliminate sources of environmental lead, children are still exposed to unsafe levels of lead from soil, dust, and water. This study investigates the effects of early-life lead exposure on alcohol consumption and anxiety-like behavior in mice. The results show that mice with early-life lead exposure consumed more alcohol and exhibited greater anxiety-like behavior. It was also found that lead exposure increased the protein content of Adenylate Cyclase-5 (AC5) in the whole brain but not in specific regions. These findings highlight the importance of understanding the mechanisms behind lead exposure and its impact on alcohol intake.

Lead content in automotive paints purchased at formal and informal outlets in Kenya

Summary: This study aimed to assess the lead (Pb) levels in automotive paints frequently used by informal spray painters. The results showed that all the paints from informal retail shops exceeded the regulatory limit of 90 ppm, with the blue paint having levels 30 times higher than the set limit. In contrast, paints from formal stores had significantly lower Pb levels than those from informal stores. Overall, high Pb levels in automotive paints used in unregulated environments could be potential sources of exposure and environmental contamination, calling for enforcement of regulatory limits and comprehensive Pb exposure assessments.

Summary: Our study determined the spatial variability of chemical element concentrations and their sources in the surficial sediments of the Boughrara Lagoon in southeastern Tunisia. The lagoon was found to be highly polluted, especially with regards to Cd, Pb, and Fe, and three potential sources of pollution were identified. Additionally, the presence of pyrite precipitation suggests the occurrence of anoxic conditions in the lagoon.

Neuroprotective Effects of Shogaol in Metals (Al, As and Pb) and High-fat diet-induced Neuroinflammation and Behavior in Mice

Summary: This study aimed to evaluate the protective effects of Shogaol (an active component of ginger) in neuroinflammation and behavioral paradigms in mice treated with toxic metals and high-fat diet. The results showed that Shogaol treatment improved metabolic profile, reduced expression of neuroinflammatory genes, and enhanced blood-brain barrier integrity. Behavioral analysis demonstrated that Shogaol treatment rescued behavioral impairments and improved anxiety and depression. Therefore, Shogaol has strong therapeutic potential and can be considered as a potential drug candidate in the future.

Health risks of heavy metals in food and their economic burden in Armenia

Elevated prelimbic cortex-to-basolateral amygdala circuit activity mediates comorbid anxiety-like behaviors associated with chronic pain

Summary: Chronic pain can lead to hyperalgesia and anxiety symptoms. This study reveals that neurons in the prelimbic cortex (PrL), which project to the basolateral amygdala (PrLBLA) and the ventrolateral periaqueductal gray (PrLl/vlPAG), exhibit increased and decreased activity, respectively, during chronic pain. Optogenetic manipulation shows that suppressing the PrL-BLA circuit reverses anxiety-like behaviors, while activating the PrL-l/vlPAG circuit reduces hyperalgesia in mice with chronic pain. Mechanistic studies suggest that elevated TNF-alpha/TNFR1 signaling in the PrL leads to increased insertion of GluA1 receptors into PrLBLA neurons and contributes to anxiety-like behaviors.

However, there is a lack of animal studies investigating the effects of low doses of lead from early stages of life to adulthood. Additionally, lead levels were elevated in blood, brain, and bone for up to six months after exposure. These findings emphasize the importance of animal models in understanding the impact of early chronic low-level lead exposure on health.

Summary: Patients with mood disorders and anxiety often show hyperresponsiveness in the hypothalamic-pituitary-adrenocortical (HPA) axis and autonomic nervous system. It has been found that p11 deficiency is associated with depression-like phenotypes. In this study, p11-deficient mice were used to investigate the role of p11 in stress responsiveness, showing increased response to stress in p11KO mice, along with hyperactivity in the HPA and SAM axes. These findings suggest that p11 deficiency may lead to increased stress reactivity and behavioral changes related to depression and anxiety.
