TGFβ1+CCR5+ neutrophil subset increases in bone marrow and causes age-related osteoporosis in male mice

Summary: TGF beta 1 induces age-related bone loss by promoting degradation of TRAF3, leading to increased numbers of TGF beta 1(+)CCR5(+) neutrophils in the bone marrow and subsequent bone loss. Inhibition of CCR5 using maraviroc can reduce TGF beta 1(+)CCR5(+) neutrophil numbers and increase bone mass in aged mice. Deletion of TGF beta RII in MPCs can also decrease TGF beta 1(+)CCR5(+) neutrophil numbers and increase bone volume. Targeting TGF beta 1(+)CCR5(+) neutrophils in the bone marrow may be a potential therapeutic approach for age-related osteoporosis.

Fetal Lc (FLc) originate from coelomic epithelium and perivascular cells, and gradually give way to adult Lc (ALc), which are considered to be derived from FLc and perivascular cells. This article summarizes the current understanding of Lc development, their cellular origins, and their functional significance in rodent and primate testes.
