Mechanism of 5S RNP recruitment and helicase-surveilled rRNA maturation during pre-60S biogenesis

Benjamin Lau et al.

Summary: This article reports nine nucleolar pre-60S cryo-EM structures from Chaetomium thermophilum, one of which clarifies the mechanism of 5S RNP incorporation into the early pre-60S. Successive assembly states then represent how helicases Dbp10 and Spb4, and the Pumilio domain factor Puf6 act in series to surveil the gradual folding of the nearby 25S rRNA domain IV. Finally, the methyltransferase Spb1 methylates a universally conserved guanine nucleotide in the A-loop of the peptidyl transferase center, thereby licensing further maturation.

Structure of nascent 5S RNPs at the crossroad between ribosome assembly and MDM2-p53 pathways

Nestor Miguel Castillo Duque de Estrada et al.

Summary: Using cryo-electron microscopy, the researchers reconstituted and determined the structure of the 5S RNP with fungal or human factors. They found that the nascent 5S rRNA associates with a nuclear import complex before developing into the 5S RNP precursor. They also elucidated the structure of an intermediate 5S RNP carrying the human ubiquitin ligase Mdm2, which explains how this enzyme can be controlled. These findings provide molecular insight into the role of the 5S RNP in ribosome biogenesis and cell proliferation.

Principles of human pre-60S biogenesis

Arnaud Vanden Broeck et al.

Summary: This study elucidates the early stages of human pre-60S assembly through a combination of human genome editing and biochemistry. The study identifies the main nucleolar assembly states and nuclear maturation states, and reveals a protein network involved in the interaction of assembly factors. The study also uncovers chemical modifications of the human 28S rRNA and identifies critical elements in generating mature large ribosomal subunits in human cells.

Dynamic association of human Ebp1 with the ribosome

Varun Bhaskar et al.

Summary: The study reports the cryo-EM structure of human Ebp1 bound to the human 80S ribosome, revealing the position and role of Ebp1 in the ribosome. Further analysis suggests that Ebp1 has the ability to rotate and may maintain interaction with the ribosome in different conformations.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.
