This interaction plays a crucial role in regulating junctional architecture and membrane properties. Cingulin b Is Required for Zebrafish Lateral Line Development Through Regulation of Mitogen-Activated Protein Kinase and Cellular Senescence Signaling Pathways

Summary: By studying the zebrafish posterior lateral line system, we discovered the important role of cingulin b in the development of auditory organs. Inhibition of cingulin b led to decreased number of neuromasts, reduced proliferative cells in the primordium, and repressed hair cell differentiation in the neuromasts. RNA-seq analysis identified MAPK signaling pathway and cellular senescence as key pathways affected by cingulin b knockdown.

Understanding and treating paediatric hearing impairment

Summary: Sensorineural hearing impairment affects 1-2 in 1000 newborns and adolescents, with over 50% being of genetic origin. Advances in clinical phenotyping, genetic diagnostics, and counselling have been made. Cochlear implants have significantly improved the situation of profoundly hearing impaired children, but there is still a major unmet clinical need for improved hearing restoration, which may be addressed through gene therapy and next generation implants in the near future.

A Novel Truncating Mutation in HOMER2 Causes Nonsyndromic Progressive DFNA68 Hearing Loss in a Spanish Family

Summary: In this study, a novel HOMER2 variant was identified in a Spanish family, expanding the mutational spectrum of this gene and contributing to clarifying the genotype-phenotype correlations of DFNA68 hearing loss. The identified mutation affects the protein structure at the same region as a previously identified mutation in a Chinese family, suggesting a common pathogenic mechanism in both populations. Age-matched patients in both families displayed similar hearing loss phenotypes.
