Polyethylene microplastics trigger cell apoptosis and inflammation via inducing oxidative stress and activation of the NLRP3 inflammasome in carp gills

Jingwen Cao et al.

Summary: Microplastics cause damage to aquatic organisms, particularly the gills which are among the first tissues to be affected. This study found that polyethylene microplastics induce inflammation, vascular remodeling, mucus cell proliferation, oxidative stress, cell apoptosis, and inflammatory immune responses in the gills. These findings provide a new target for preventing toxin-induced gill injury from polyethylene microplastics.

TBBPA induced ROS overproduction promotes apoptosis and inflammation by inhibiting autophagy in mice lung

Yanhe Zhang et al.

Summary: Tetrabromobisphenol A (TBBPA), a non-degradable environmental pollutant, is released into the air during the production, use, and recycling of plastic products. Respiratory exposure is the main route of TBBPA inhalation. However, research on the respiratory system damage caused by TBBPA is limited. This study aimed to investigate the mechanism of TBBPA toxicity to the lungs. Results showed that exposure to TBBPA led to inflammatory cell infiltration and tissue structure damage in the lungs of mice. Increased levels of ROS and MDA, as well as inhibition of T-AOC, GSH-Px, CAT, and SOD activities, were observed in lung tissue with TBBPA exposure. The expression of autophagy-related factors Beclin-1, P62, LC3-II, ATG5, and ATG7 decreased. Activation of the NF kappa B/TNF-alpha pathway indicated inflammation. Increased expression of Bax, caspase3, caspase7, caspase 9, decreased expression of Bcl-2, and activation of the apoptosis pathway were observed. The autophagy inducer rapamycin could reverse the adverse effects of inflammation and apoptosis. Together, TBBPA inhibits autophagy-induced pneumonia and apoptosis by overproducing ROS.

Polystyrene microplastics induce endoplasmic reticulum stress, apoptosis and inflammation by disrupting the gut microbiota in carp intestines

Fuhan Wang et al.

Summary: Microplastics have been identified as a widespread pollutant in nature, causing various diseases in carp. A model of microplastic ingestion was successfully established, and the gut microbiota was analyzed. The results showed a reduction in specific bacteria after treatment with PS-MPs, and related intestinal flora disorders were found. Ingestion of PS-MPs resulted in intestinal tissue damage, apoptosis, and increased inflammation. The occurrence of ERS and apoptosis in carp intestines was confirmed. These findings indicate that the accumulation of PS-MPs in the aquatic environment can disrupt the carp gut microbiota and induce ERS, apoptosis, and inflammation in the intestinal tissue.

Polystyrene microplastics induce myocardial inflammation and cell death via the TLR4/NF-?B pathway in carp

Qirui Zhang et al.

Summary: This study constructed a carp polystyrene microplastics (PS-MPs) exposure model to investigate the damage and mechanism of PS-MPs exposure to carp myocardial tissue. The results showed that PS-MPs exposure induced inflammation, apoptosis, and necrosis in carp myocardial tissue and cardiomyocytes. Furthermore, PS-MPs exposure significantly increased the expression of TLR4 pathway-related factors and the NF-kappa B pathway and inflammation-related factors in a dose-dependent manner. These findings provide essential insights into the toxicological studies on PS-MPs and have important implications for aquaculture.

Zinc deficiency causes oxidative stress, endoplasmic reticulum stress, apoptosis and inflammation in hepatocytes in grass carp

Jie Cui et al.

Summary: A lack of zinc in freshwater environments causes slow growth, malnutrition, and abnormal biological functions in organisms. In this study, a zinc deficiency model of grass carp hepatocytes was established using TPEN. The results showed increased LDH, AST, ALT, and AKP activities, as well as ROS levels and decreased antioxidant enzyme activities in a zinc-deficient environment. RT-PCR and ELISA demonstrated increased expression levels of genes related to endoplasmic reticulum stress, apoptosis, and inflammation. These findings suggest that zinc deficiency induces inflammation and apoptosis in grass carp hepatocytes in the presence of calcium, highlighting the adverse effects of zinc deficiency in freshwater environments.

Microplastics induced inflammation and apoptosis via ferroptosis and the NF-KB pathway in carp

Tianchao Xu et al.

Summary: In this study, a model of carp exposed to microplastics was successfully established. It was observed that exposure to microplastics caused damage to the intestinal mucosal surface and an increase in goblet cells. The study also found that microplastic exposure led to lymphocyte proliferation and aggregation in the intestinal wall, resulting in cellular apoptosis. The activation of NF-KB pathway and ferroptosis induced by lipid peroxidation were identified as the potential mechanisms underlying microplastic-induced intestinal damage.

The dual role of p62 in ferroptosis of glioblastoma according to p53 status

Fanen Yuan et al.

Summary: This study reveals a dual role of p62 in regulating ferroptosis in glioblastoma (GBM) depending on the status of p53. P62 overexpression promotes ferroptosis and inhibits SLC7A11 expression in p53 mutant GBM, while attenuates ferroptosis and promotes SLC7A11 expression in p53 wild-type GBM. The mutation status of p53 is a crucial factor in determining the therapeutic response to p62-mediated ferroptosis-targeted therapies in GBM.

KIF5A-dependent axonal transport deficiency disrupts autophagic flux in trimethyltin chloride-induced neurotoxicity

Summary: TMT exposure impairs autophagic flux and disrupts lysosomal function, leading to neuronal death. KIF5A plays a crucial role in TMT-induced neurotoxicity by affecting axonal transport and autophagic clearance. Overexpression of KIF5A can alleviate TMT-induced neurotoxicity by restoring lysosomal function.

Asprosin promotes β-cell apoptosis by inhibiting the autophagy of β-cell via AMPK-mTOR pathway

Summary: The study demonstrated that asprosin induces apoptosis in beta-cells by inhibiting autophagy. High glucose and asprosin both lead to increased apoptosis and altered expression of certain proteins in MIN6 cells, affecting the AMPK-mTOR signaling pathway. Activation of the AMPK pathway by AICAR partially reverses the effects of asprosin on autophagy and apoptosis in beta-cells.
