Summary: Non-alcoholic fatty liver disease (NAFLD) is the most common liver disease worldwide, and no specific medicine has been approved for its treatment. The study found that TRIM59 was highly expressed in NAFLD tissues and its inhibition could suppress steatosis and inflammation in NAFLD, while its overexpression had the opposite effects. TRIM59 was shown to interact with GPX4 and enhance its ubiquitination, and GPX4 overexpression could reverse the pathogenic effects of TRIM59 in NAFLD.

Nonalcoholic fatty liver disease and obstructive sleep apnea in women with polycystic ovary syndrome

Summary: Polycystic ovary syndrome (PCOS) is frequently associated with nonalcoholic fatty liver disease (NAFLD) and obstructive sleep apnea. Women with PCOS have a higher risk of NAFLD compared to healthy weight-matched controls, independent of body mass index. Insulin resistance and hyperandrogenemia play a central role in the development of NAFLD. Timely diagnosis of NAFLD is important as it can progress to nonalcoholic steatohepatitis and advanced liver fibrosis, leading to liver-related mortality. NAFLD is also associated with increased risks of type 2 diabetes, cardiovascular events, overall mortality, and extrahepatic cancers.

It is found that enhanced expression of TRIM67 leads to hepatic inflammation, disruption of lipid metabolic homeostasis, and promotion of NAFLD progression induced by obesity. Furthermore, the transcription coactivator PGC-1 alpha is implicated in the response of hepatic TRIM67 to obesity.
