Twist2-driven chromatin remodeling governs the postnatal maturation of dermal fibroblasts

Jin Yong Kim et al.

Summary: Dermal fibroblasts lose stem cell potency after birth, hindering regenerative healing. This study reveals that postnatal maturation of papillary fibroblasts (PFs) is driven by Twist2-mediated remodeling of chromatin accessibility. The loss of regenerative ability in postnatal PFs is associated with decreased H3K27ac levels and chromatin accessibility. Inhibition of histone deacetylase delays chromatin remodeling, preserving the regenerative ability of postnatal PFs.

CBP/p300 and HDAC activities regulate H3K27 acetylation dynamics and zygotic genome activation in mouse preimplantation embryos

Meng Wang et al.

Summary: Epigenome reprogramming after fertilization is crucial for early embryonic development. The chromatin marker H3K27ac undergoes three waves of global transitions during this process. CBP/p300 acetyltransferase deposits H3K27ac and promotes chromatin opening, while HDAC prevents premature expression of developmental genes.

Dynamic Transcriptional and Epigenetic Changes Drive Cellular Plasticity in the Liver

Allyson J. Merrell et al.

Summary: Hepatocytes undergo extensive chromatin and transcriptional changes during biliary reprogramming, resulting in altered gene expression profiles including robust induction of biliary genes and weaker repression of hepatocyte genes. Single-cell analysis revealed that Smad4 mutation can significantly increase reprogramming.
