Reassessing the embryonic origin and potential of craniofacial ectomesenchyme

Peter Fabian et al.

Summary: Of all the cell types arising from the neural crest, ectomesenchyme is unique as it generates connective tissue and skeletal cell types in common with mesoderm. The origin and potential of this ectoderm-derived mesenchyme is still debated, and recent studies are exploring intrinsic and extrinsic cues in determining its potential and its influence on craniofacial repair.

Gene regulatory network from cranial neural crest cells to osteoblast differentiation and calvarial bone development

Junguang Liao et al.

Summary: This article summarizes the important role of the cranial neural crest (CNC) in calvarial bone development, including discussing the potential cell fate of CNC and the gene regulatory network that controls this potential. Furthermore, the study has found a close correlation between CNC patterning disorders and clinical birth defects and diseases. The article emphasizes the potential regenerative alternative based on CNC patterning and genetic molecules for treating clinical disorders.

AP-2α and AP-2β cooperatively function in the craniofacial surface ectoderm to regulate chromatin and gene expression dynamics during facial development

Eric Van Otterloo et al.

Summary: The facial surface ectoderm plays a vital role in the development of cranial neural crest cell populations by providing signals for growth, patterning, and morphogenesis. This study shows that two members of the AP-2 transcription factor family, AP-2 alpha and AP-2ss, are important for normal craniofacial development and signaling. The loss of these transcription factors leads to clefts in the upper face and mandible, as well as fusion of the upper and lower jaws in the hinge region.

Role of Wnt-signaling inhibitors DKK-1 and sclerostin in bone fragility associated with Turner syndrome

M. Chiarito et al.

Summary: This study aimed to deepen the understanding of skeletal fragility in girls with Turner syndrome (TS) by evaluating the serum levels of Dickkopf-1 (DKK-1) and sclerostin, as well as the percentage of circulating osteoblast precursors (OCPs). The results showed that TS patients had higher levels of sclerostin and DKK-1, which may be related to hormonal replacement therapy (HRT) and reduced bone formation markers, as well as increased bone resorption activity.

Conditional Deletion of AP-2β in the Periocular Mesenchyme of Mice Alters Corneal Epithelial Cell Fate and Stratification

Haydn Walker et al.

Summary: The cornea is a crucial anterior eye structure specialized for vision, with the corneal epithelium and stroma derived from POM influencing its development and structure. Deletion of AP-2β leads to corneal epithelial abnormalities, pointing to a potential regulatory role of POM-derived stromal signaling in cell fate and stratification in the cornea.

A bibliometric analysis of Wnt signaling pathway: from the top-100 cited articles to emerging trends

Jia-Qi Xu et al.

Summary: Research on Wnt signaling pathway from 2000 to 2009 mainly focused on cancer, embryo development, and cytoplasm signal transduction. However, emerging subtopics such as osteogenesis, immune, apoptosis, autophagy, microRNA, and cancer stem cell have been gaining interest in recent years. The association of Wnt pathway with diseases was further revealed through integrated investigation on the network of Wnt signaling.
