Behavioral, Neurochemical and Developmental Effects of Chronic Oral Methylphenidate: A Review

Summary: The majority of animal studies on methylphenidate (MP) use methods such as intraperitoneal injections, subcutaneous injections, or the oral gavage route. However, the most clinically relevant route is oral administration. Intraperitoneal injections provide an immediate and maximum dose, but only represent a limited window of the drug's effects. Subcutaneous injections do not accurately reflect oral exposure, and the oral-gavage method has adverse effects and may be stressful for animals. The use of a two-bottle drinking method allows for free consumption of MP and more accurately mirrors human treatment, enabling the study of MP's effects on development, behavior, neurochemistry, and brain function.

Recent studies have identified a significant overlap of dopaminergic gene risk polymorphisms in depressed and schizophrenic individuals. ADHD, PTSD, and spectrum disorders have also been linked to the neurogenetic and psychological underpinnings of RDS. It is believed that RDS may be a foundational aspect of species evolution and survival, influenced by multiple neurotransmitters and polymorphic loci.

Summary: Since 1990, there has been a surge in genetic candidate association studies, including GWAS, following our laboratory's publication in JAMA on the association of the DRD2 Taq A1 allele and severe alcoholism. Blum's group developed the Genetic Addiction Risk Severity (GARS) test, which consists of ten genes and eleven associated risk alleles, to accurately identify those at risk for Alcohol Use Disorder (AUD). Our statistical analysis of studies published from 1990 to 2021 validated the selection of these risk alleles measured by GARS, with significant correlations found for DRD2, DRD3, DRD4, DAT1, COMT, OPRM1, and 5HTT. While most of the research related to GARS is derived from our laboratory, independent research is encouraged to confirm our findings.
