Experimental Models of In Vitro Blood-Brain Barrier for CNS Drug Delivery: An Evolutionary Perspective

Summary: Central nervous system (CNS) disorders are a major global health burden, yet the approval rate for new therapies against these disorders is low compared to other diseases. The lack of reliable and efficient in vitro blood-brain barrier (BBB) models hinders the development of successful therapy for CNS disorders. This review discusses the major in vitro BBB models, including 2D transwell models, 3D organoid models, and microfluidic models, for exploring the barrier properties of the cerebral vasculature. Although microfluidic models better recapitulate the in vivo properties of BBB, significant gaps still exist for their use in predicting the performance of neurotherapeutics.

The blood-brain barrier: structure, regulation, and drug delivery

Summary: The blood-brain barrier (BBB) is a natural protective membrane that prevents toxins and pathogens in the blood from entering the central nervous system (CNS). However, this barrier hinders the delivery of drugs to the brain, leading to low therapeutic efficacy and increased side effects. Recent advances in materials science and nanotechnology have provided advanced materials with customized structures and properties for targeted drug delivery. This review summarizes the physiological structure of the BBB and different cells contributing to its formation, as well as various strategies to enhance BBB crossing and regulate its permeability.

Function and therapeutic value of astrocytes in neurological diseases

Summary: This review discusses astrocytes, a type of glial cell that is involved in various neurological diseases. Astrocytes regulate important processes in the central nervous system, such as glutamate and ion homeostasis, cholesterol and sphingolipid metabolism, and response to environmental factors. The review highlights the potential of targeting astrocytes for therapeutic intervention, including manipulating their metabolism and targeting transporters and receptors on astrocytes. The review also proposes a new nomenclature for astrocyte subsets to better understand their roles in neurological diseases.

Molecular dynamics simulations were used in this study to investigate the interactions between benzimidazolyl-urea and a phospholipid bilayer, showing that the permeation process is hindered by high energy barriers, potentially leading to clustering and pore formation with increasing concentration.

Microglia Specific Drug Targeting Using Natural Products for the Regulation of Redox Imbalance in Neurodegeneration

Summary: Microglia, an innate immune cell of the brain, plays a crucial role in regulating neurogenesis, immunological surveillance, redox imbalance, and cognitive functions. Under normal conditions, it maintains homeostasis by producing cytokines, but in neurodegenerative diseases, it can switch to an activated phenotype releasing pro-inflammatory cytokines, leading to neuroinflammation and redox imbalance. Targeting microglia pharmacologically may offer a promising strategy to manage neuroinflammation and oxidative stress in diseases like Alzheimer's and Parkinson's.

Alkylsaccharide excipients, such as dodecyl maltoside, have shown to be nonirritating and effective in enhancing systemic bioavailability, and are used in FDA-approved intranasal formulations for migraine and epilepsy patients.
