Continuous crystallization and its potential use in drug substance Manufacture: A review

Ayse Eren et al.

Summary: In recent years, there has been a significant increase in interest in continuous manufacturing of active pharmaceutical ingredients (API's) in the pharmaceutical industry. This has led to extensive research on continuous processing, particularly in the area of continuous crystallization. The majority of this research has focused on the Mixed Suspension Mixed Product Removal (MSMPR) crystallizer, which will be described in this review along with its applications in continuous API manufacturing.

A Comparative Investment Analysis of Batch Versus Continuous Pharmaceutical Manufacturing Technologies

Clifford Rossi

Summary: This study examines the risks and economics of investing in continuous manufacturing versus conventional batch manufacturing for oral solid dosage pharmaceutical products in the USA and abroad. The results suggest that the lower costs associated with continuous manufacturing technology make it more attractive for both brand and generic companies. The study also shows that US tax policy has a significant impact on pharmaceutical companies' manufacturing investment decisions.

Design and characterization of Kenics static mixer crystallizers

Kiran Mathew Thomas et al.

Summary: This study evaluated the performance of a tubular crystallizer with gaps between the Kenics type mixing elements and compared it with a standard Kenics static mixer and a hollow tubular crystallizer. The gapped Kenics static mixer crystallizer showed advantages in terms of mixing length, shear rate, residence time distribution, pressure drop, and segregation or settling of crystals. Additionally, its crystallization performance, in terms of desupersaturation profile and mean crystal size, was similar to that of the standard Kenics static mixer crystallizer. The advantages of the gapped Kenics static mixer crystallizer, such as lower pressure drop and minimization of crystal settling, allow for lower flow rates and longer residence times under plug flow conditions, which is beneficial for continuous crystallization.

Continuous Crystallization Processes in Pharmaceutical Manufacturing: A Review

Jaka Orehek et al.

Summary: This scientific paper provides an overview of continuous solution crystallization in the pharmaceutical industry, covering topics such as comparisons between different crystallizers, solid form structuring, and online process analytical technology. It discusses mechanistic multiscale modeling, model-based predictive control, and the introduction of new continuous systems like the coiled flow inverter crystallizer. Continuous crystallization is highlighted for its efficiency, on-demand production, and potential for optimization and intensification.

Influence of Buoyancy Effects on the Mixing Process and RTD in a Side-Injection Reactor Equipped with Static Mixers

Jody Albertazzi et al.

Summary: In this study, the mixing and RTD of two miscible liquids with different densities and viscosities in a side-injection tubular reactor equipped with Sulzer Static Mixers were investigated through BANS-based CFD simulations. The results show that with proper configurations, radial mixing and plug flow behavior can be approximated well, even when there are large differences in densities. Graphics for quickly estimating the maximum mixing length and the corresponding Pe(-1) value as a function of the Re number were also obtained.

CFD Analysis of a Kenics Static Mixer with a Low Pressure Drop under Laminar Flow Conditions

Baggie W. Nyande et al.

Summary: Kenics static mixers achieve enhanced mixing efficiency at lower pressure drops by introducing gaps between the mixing elements. The gaps enhance mixing effects at the entrances and exits of the mixing elements, providing a compromise between the required mixing length and pressure drop. The presence of gaps allows for a narrower residence time distribution and suppression of Taylor dispersion, improving overall mixing performance.

Mixing Efficiency and Residence Time Distributions of a Side-Injection Tubular Reactor Equipped with Static Mixers

Jody Albertazzi et al.

Summary: The study investigates radial mixing and residence time distributions in a side-injected tubular reactor equipped with static mixers using Computational Fluid Dynamics. The findings show that the reactor can efficiently satisfy plug flow conditions even at low Reynolds numbers. Operative diagrams are determined to predict reactor behavior.
