Rucaparib cocrystal: Improved solubility and bioavailability over camsylate

Summary: In this study, new solid forms of Ruc were prepared through cocrystallization to improve its oral exposure. The cocrystals of Ruc-Thp MH, Ruc-Mal, and Ruc-Emal were obtained and characterized. It was found that Ruc-Thp MH had higher solubility and bioavailability compared to Ruc-Cam.

The influence of different carbonate ligands on the hydrolytic stability and reduction of platinum(iv) prodrugs

Summary: This study synthesized six mono-carbonated Pt(iv) complexes with axial carbonate ligands of various electron-withdrawing abilities, and prepared corresponding mono-carboxylated analogues as references to investigate the differences in hydrolysis and reduction rates. It was found that mono-carbonated Pt(iv) complexes are less stable and reduced faster compared to mono-carboxylated ones, and the hydrolysis and reduction profiles depend on both the electron-withdrawing ability of the carbonates and the nature of the opposite axial ligands.

Crystal Engineering of Pharmaceutical Cocrystals in the Discovery and Development of Improved Drugs

Summary: Crystal engineering is a discipline that started in the 1970s, focusing on the study of topochemical reactions in the solid state. One important application of crystal engineering is in the design of pharmaceutical cocrystals, which can systematically tune the physicochemical properties of drugs without altering their molecular structure. This review traces the development of supramolecular synthons as robust hydrogen bond patterns for pharmaceutical cocrystal design, and discusses recent advancements in the field that have led to the launch of several drugs with improved efficacy and bioavailability. The importance of pharmaceutical cocrystals lies in their ability to bridge the gap between drug discovery and development, and their potential to provide high-value pharmaceuticals with economic and environmental benefits.

The first marine dual-drug cocrystal of cytarabine with 5-fluorouracil having synergistic antitumor effects shows superior biopharmaceutical peculiarities by oral administration

Summary: This research highlights the advantages of cocrystallization technique in improving the in vitro/vivo properties of the marine drug cytarabine (ARC) and fills the research gap of marine pharmaceutical cocrystals with synergistic antitumor effects. The study successfully designs and assembles a dual-drug cocrystal containing ARC and the antitumor drug 5-fluorouracil (FU), named ARC-FU. The accurate structure of the cocrystal is resolved through X-ray diffraction and other methods. The results demonstrate that the cocrystal consists of ARC and FU with a molar ratio of 1:1, and the structure and aggregation model of the cocrystal play a crucial role in improving the in vitro/vivo properties. The in vitro studies show reduced solubility and increased permeability for the cocrystal, in accordance with theoretical predictions. Importantly, the improved in vitro characteristics translate to in vivo pharmacokinetic optimization, including extended residence time and enhanced bioavailability. Furthermore, the ARC-FU cocrystal exhibits synergistic antitumor effects with lower IC50. This research not only provides a novel crystalline form with forward-looking development value for ARC, but also paves the way for the development of synergistic antitumor pharmaceutical cocrystals with marine characteristics.

Slow-Release Drug-Drug Cocrystals of Oxaliplatin with Flavonoids: Delaying Hydrolysis and Reducing Toxicity

Summary: Two novel drug-drug cocrystals of Oxaliplatin were synthesized and characterized, showing reduced release rate and delayed hydrolysis compared to Oxaliplatin. Theoretical analysis revealed that hydrogen bonds were essential for reducing the dissolution rate of the cocrystals. Cell toxicity evaluation suggested that one cocrystal may have higher safety, while the other may have more significant effects on inhibiting cancer cells.

Summary: Pharmaceutical cocrystals and salts are extensively studied for their ability to modify the physicochemical properties of active pharmaceutical ingredients. In this study, different forms of olanzapine were discovered through solvent drop grinding and ball milling. These new solid phases all followed the Delta pKa rule and showed increased solubility due to strong hydrogen bonding interactions.
