Esophageal Cancer Stem-like Cells Resist Ferroptosis-Induced Cell Death by Active Hsp27-GPX4 Pathway

Chen-Chi Liu et al.

Summary: Cancer stem cells (CSCs) in esophageal squamous cell carcinoma are more susceptible to ferroptosis-inducing agents, but can be rescued from cell death by activating Hsp27 to upregulate GPX4. Targeting Hsp27 or GPX4 to block this protective mechanism against ferroptosis may be a potential treatment strategy for eradicating CSCs in this type of cancer.

Trigonelline inhibits Nrf2 via EGFR signalling pathway and augments efficacy of Cisplatin and Etoposide in NSCLC cells

Chandrani Fouzder et al.

Summary: The study demonstrated that Trigonelline inhibits Nrf2 activation and its nuclear translocation by blocking the EGFR signaling pathway, enhancing the responsiveness of NSCLC cells to Cisplatin and Etoposide without visible cytotoxicity to normal lung epithelial cells. It showed strong synergism in combination with Cisplatin/Etoposide at lower concentrations than the IC50 values of these drugs, indicating its potential as a promising choice for lung cancer therapy.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.
