Bone Microenvironment-Mimetic Scaffolds with Hierarchical Microstructure for Enhanced Vascularization and Bone Regeneration

Summary: This study develops a bone microenvironment-mimicking scaffold with microchannel networks and nanofibrous structures using 3D printing and thermally induced phase separation techniques. The scaffold promotes cells migration and nutrient transportation. Cell experiments show that the scaffold promotes angiogenesis and osteogenesis, and enhances the angiogenic activity of osteoblasts through the activation of specific pathways.

By employing bioorthogonal clicking, antimicrobial peptide (AMP) and osteogenic growth peptide (OGP) were precisely located on the azido-modified PEEK surfaces, resulting in a dual-effect of host defense and tissue repair.

Effect of hydroxyapatite content and particle size on the mechanical behaviors and osteogenesis in vitro of polyetheretherketone-hydroxyapatite composite

Summary: PEEK-HA composites show impaired tensile/bending strength and elongation at break with increasing HA content, but improved modulus. The compressive strength and modulus of PEEK-HA are enhanced with increasing HA amount. Smaller HA particle size in PEEK-HA leads to higher tensile/bending performance and lower compressive properties. In vitro osteogenesis tests reveal that higher HA content and smaller particle size result in more evenly flattened cells with increased cytoplasmic processes. The PEEK-HA composite with 20 wt% HA content and 20 nm particle size shows optimal osteogenesis performance while maintaining sufficient mechanical properties.
