Ex Vivo and In Vivo Gene Therapy for Mucopolysaccharidoses: State of the Art

Giulia Consiglieri et al.

Summary: Available treatment options for MPSs have limitations, and improved therapeutic approaches are needed. Gene therapy shows potential in treating MPSs, particularly in skeletal and CNS manifestations. Significant progress has been made in ex-vivo and in-vivo gene therapy for MPSs, leading to the marketing authorization of medicinal product Libmeldy. Clinical trials for MPSIH and MPSIIIA have shown promising results, although longer follow-up is needed. Gene-corrected cells can cross-correct nonhematopoietic cells, potentially improving skeletal manifestations. Gene therapy has advantages over allo-HSCT, but further long-term studies are necessary to confirm efficacy.

3D inkjet printed self-propelled motors for micro-stirring

Piyush Kumar et al.

Summary: In this study, millimeter-scale self-propelled motors (SPMs) were successfully fabricated using 3D reactive inkjet printing. Different geometric shapes and propulsion mechanisms were compared, and the results showed that surface tension propulsion resulted in faster velocities with smooth deceleration, making it suitable for enhancing the sensitivity of diffusion-rate limited biomedical assays.

Scaffold-free cell-based tissue engineering therapies: advances, shortfalls and forecast

Andrea De Pieri et al.

Summary: Cell-based scaffold-free therapies aim to create in vitro organotypic three-dimensional tissue-like surrogates, but they still face challenges in terms of cost and technology transfer in clinical applications. While automation systems have been implemented, achieving efficient scaffold-free organogenesis remains difficult.

3D Cell Culture: Recent Development in Materials with Tunable Stiffness

Desiree Baruffaldi et al.

Summary: Three-dimensional cell culture systems are believed to better simulate physiological conditions compared to traditional two-dimensional systems. Mechanosensing plays a crucial role in controlling various cell functions, and alterations in extracellular matrix structure due to pathologies can impact tissue stiffness. Development of platforms with modulable stiffness may enhance understanding of cell behavior under mechanical stress stimuli.
