Predictive modeling as a tool to assess polymer-polymer and polymer-drug interactions for tissue engineering applications

Summary: The success of tissue engineering scaffolds for wound healing depends on their physicochemical properties and the addition of small molecules. In this study, machine learning methods were applied to classify and predict the miscibility of different polymer blends. The random forest classifier performed the best, and a multiple linear regression model was used to analyze polymer-drug interactions. The results showed that UV absorbance of polymer-drug solutions could be correlated with regression models.

Highly Retina-Permeating and Long-Acting Resveratrol/Metformin Nanotherapeutics for Enhanced Treatment of Macular Degeneration

Summary: We developed a nanomedicine strategy to create nanotherapeutics with high retinal permeability and sustained bioactive delivery. The nanotherapeutics were designed through aminolysis of resveratrol-encapsulated polycaprolactone nanoparticles, followed by the formation of amide linkages with a transacting activator of transcription cell penetrating peptide and metformin. In vitro and in vivo studies demonstrated the effectiveness of the nanotherapeutics in improving retinal permeability and targeting retinal diseases.

Tyrosine kinase SRC-induced YAP1-KLF5 module regulates cancer stemness and metastasis in triple-negative breast cancer

Summary: The study reveals that SRC regulates cancer stemness and metastasis in triple-negative breast cancer (TNBC) through a YAP1 tyrosine phosphorylation-dependent YAP1-KLF5 oncogenic module. Activation of SRC increases cancer stemness, tumor cell growth, and metastasis, while inhibition of SRC kinase reduces them. The interaction between YAP1 and KLF5, induced by SRC-mediated YAP1 tyrosine phosphorylation, promotes TEAD-mediated transcriptional program independently of canonical Hippo kinases, leading to enhanced cancer stemness and metastasis.

Poly(l-Histidine)-Mediated On-Demand Therapeutic Delivery of Roughened Ceria Nanocages for Treatment of Chemical Eye Injury

Summary: Development of topical bioactive formulations capable of overcoming low bioavailability of conventional eye drops is crucial for effective treatment of ocular chemical burns. In this study, a nanomedicine strategy utilizing surface roughness-controlled ceria nanocages (SRCNs) and poly(l-histidine) surface coatings is proposed to achieve multiple bioactive functions of therapeutic nanocarriers, enhance transport across corneal epithelial barriers, and enable on-demand release of dual drugs. The nanoformulation was found to effectively reduce corneal wound areas, attenuate abnormal blood vessels, and restore corneal transparency, demonstrating great potential for ocular pharmacology and tissue regenerative medicine.

Structural characterization of a P-selectin and EGFR dual-targeting fucoidan from Sargassum fusiforme
