Emerging functions and clinical applications of exosomal microRNAs in diseases

Soudeh Ghafouri-Fard et al.

Summary: Exosomes are extracellular vesicles that transfer biomolecules and facilitate cell-cell communication. The content, particularly miRNA, in exosomes reflects disease-specific patterns and can be used as diagnostic and prognostic markers. Exosomal miRNAs enter recipient cells and regulate gene expression. They have great potential in detecting disorders, especially cancers. However, standardization, large-scale clinical studies, and consistent experimental settings are challenges that need to be addressed.

Review on Strategies and Technologies for Exosome Isolation and Purification

Jiaci Chen et al.

Summary: Exosomes, as nano-sized extracellular vesicles, play an important role in cell-specific material transfer and have potential applications in prognosis, diagnosis, and treatment. Existing exosomal separation techniques have limitations in terms of complexity, time consumption, and low purity, highlighting the need for efficient and simple isolation methods. Microfluidic chips have emerged as a promising technology for exosome isolation. A comprehensive review covering exosomal properties, conventional technologies, emerging isolation strategies, and large-scale production of engineered exosomes is essential.

Neuronal and Astrocytic Extracellular Vesicle Biomarkers in Blood Reflect Brain Pathology in Mouse Models of Alzheimer's Disease

Francheska Delgado-Peraza et al.

Summary: The study found that biomarkers in circulating NEVs and AEVs reflect their brain levels across multiple AD mouse models, supporting their potential use as a liquid biopsy for neurological disorders. This demonstrates a potential for utilizing EV proteins as AD biomarkers and emphasizes the importance of further research in this area.

Decoding distinctive features of plasma extracellular vesicles in amyotrophic lateral sclerosis

Laura Pasetto et al.

Summary: The study analyzed plasma-derived EVs of ALS patients and controls, as well as ALS mouse models. Results showed distinct size distribution and lower HSP90 levels in the EVs of ALS patients and mouse models compared to controls. Additionally, distinguishing fast and slow disease progressors based on EV size distribution and cyclophilin A levels may offer a new approach for patient stratification.

Formation of a protein corona on the surface of extracellular vesicles in blood plasma

Eszter A. Toth et al.

Summary: This study found that a protein corona is formed around EVs in blood plasma, with shared EV corona proteins identified. These results help explain the commonly reported plasma protein 'contamination' of EV preparations and offer a new perspective for EV research.
