Neural stem/progenitor cell therapy for Alzheimer disease in preclinical rodent models: a systematic review and meta-analysis

Zijing Zhou et al.

Summary: This study evaluated the effectiveness of neural stem/progenitor cell (NSPC) therapy in rodent models of Alzheimer's disease (AD). The results showed that NSPC therapy significantly improved cognitive function and reduced pathological features (Aβ deposition and phosphorylated tau levels). Additionally, nanoformulation and genetic modification enhanced the effects of NSPC therapy. Further animal trials and clinical trials are needed to validate the efficacy of this treatment.

Insight into the role of adult hippocampal neurogenesis in aging and Alzheimer's disease

Peng Chen et al.

Summary: Alzheimer's disease is a common form of dementia that significantly impacts the quality of life for the elderly. Hippocampal dysfunction plays a pivotal role in neurodegeneration in Alzheimer's patients and understanding the impact of neurogenesis on aging and cognitive deficits is crucial for advancing research in this field.

Investigation of the Molecular Role of Brain-Derived Neurotrophic Factor in Alzheimer's Disease

Pragya Girotra et al.

Summary: BDNF, a member of the neurotrophin family, plays a crucial role in normal neural maturation and is associated with altered levels in patients with AD. AD is a progressive and form of dementia that leads to cognitive decline impacting thinking, memory, and behavior. Pathologically, AD involves the degeneration of neurons in the cerebral cortex with the accumulation of A beta and formation of NFTs, affecting neuron signaling and leading to dementia.

Hippocampal Iron Accumulation Impairs Synapses and Memory via Suppressing Furin Expression and Downregulating BDNF Maturation

Yating Zhang et al.

Summary: This study demonstrates that hippocampal iron overload leads to reduced furin expression, which impairs the maturation of brain-derived neurotrophic factor and downregulates synaptogenesis-related proteins, resulting in cognitive decline. Iron chelation or furin overexpression can restore these abnormalities and improve cognitive function. Therefore, hippocampal iron accumulation and the inhibition of furin may play an important role in iron-mediated synaptic damage and memory loss in Alzheimer's disease.

Astrocyte-derived hepcidin controls iron traffic at the blood-brain-barrier via regulating ferroportin 1 of microvascular endothelial cells

Linhao You et al.

Summary: Brain iron dysregulation associated with aging is closely related to motor and cognitive impairments in neurodegenerative diseases. The study reveals that astrocytes regulate iron transport into the brain by controlling the expression of FPN1 in brain microvascular endothelial cells (BMVECs) through the secretion of hepcidin. The results provide potential therapeutic targets for iron dysregulation-related neurological diseases.

Treadmill Exercise Alleviates Brain Iron Dyshomeostasis Accelerating Neuronal Amyloid-β Production, Neuronal Cell Death, and Cognitive Impairment in Transgenic Mice Model of Alzheimer's Disease

Dong-Hun Choi et al.

Summary: Brain iron levels increase with age, and abnormal iron metabolism may be associated with Alzheimer's disease pathology. Studies suggest that physical exercise can mitigate abnormal brain iron regulation in AD, reducing cognitive decline and neuronal cell death risk.

Evidences for Adult Hippocampal Neurogenesis in Humans

Elena P. Moreno-Jimenez et al.

Summary: The rodent hippocampus generates new neurons throughout life, but direct evidence of adult neurogenesis in humans remains elusive. Research indicates that adult hippocampal neurogenesis may persist until the 10th decade of human life, with marked impairments in patients with Alzheimer's disease. Methodological aspects in processing and analyzing postmortem human brain samples may limit the detection of markers of adult hippocampal neurogenesis, highlighting the need for strict controls in human studies.

Ferroptosis: mechanisms and links with diseases

Hong-fa Yan et al.

Summary: Ferroptosis is a form of cell death that is iron-dependent and involves accumulation of lethal lipid species from lipid peroxidation. This review discusses the regulatory mechanisms of ferroptosis and its connections with iron, lipid, and cysteine metabolism pathways. Additionally, it explores the role of ferroptosis in diseases such as cancer, ischemia/reperfusion, Alzheimer's disease, and Parkinson's disease, and evaluates the potential therapeutic applications of ferroptosis inhibitors in clinical settings.
