A sleep-active basalocortical pathway crucial for generation and maintenance of chronic pain

Summary: Poor sleep is associated with the risk of developing chronic pain. In a mouse model of neuropathic pain, cholinergic neurons in the basal forebrain are increasingly active during nonrapid eye movement (NREM) sleep. These neurons directly activate vasoactive intestinal polypeptide-expressing interneurons in the primary somatosensory cortex (S1), causing pain. The hyperactivity of these neurons is caused by inputs from the parabrachial nucleus (PB) driven by the injured peripheral nerves.

Contralateral S1 function is involved in electroacupuncture treatment-mediated recovery after focal unilateral M1 infarction

Summary: Acupuncture at acupoints Baihui (GV20) and Dazhui (GV14) has been shown to promote functional recovery after stroke. In this study, the researchers found that after unilateral ischemic infarction of the primary motor cortex, the blood perfusion and neuronal interaction between the contralateral primary sensory cortex (S1) and primary motor cortex (M1) were impaired. However, electroacupuncture (EA) treatment restored the impaired intrinsic neuronal excitability and activity. Furthermore, the effectiveness of EA treatment was inhibited after virus-mediated neuronal ablation of the contralateral S1, suggesting the importance of neuronal activity in contralateral S1 for EA-mediated recovery after focal M1 infarction. The study provides insight into the mechanism of EA treatment for unilateral cerebral infarction involving the Si-M1 circuit.
