Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Protein disulfide isomerase a4 promotes lung cancer development via the Stat3 pathway in stromal cells

Tzung-Yan Chen et al.

Summary: The expression of Pdia4 in lung cancer was found to be negatively correlated with patient survival. Pdia4 was expressed at a low level in stromal cells, and its expression was up-regulated by tumor cell-derived stimuli. Host Pdia4 was shown to promote lung cancer development in mice via cancer stroma, and it positively regulated the number and immunosuppressive function of stromal cells.

HOXA10 promote pancreatic cancer progression via directly activating canonical NF-κB signaling pathway

Jiao Li et al.

Summary: Although HOXA10 has been shown to play an important role in pancreatic development, its involvement in pancreatic ductal adenocarcinoma (PDAC) progression remains unclear. In this study, we found that HOXA10 expression increased in PDAC with higher tumor stage and poor patient survival. HOXA10 promoted PDAC cell proliferation, colony formation, and xenograft growth by activating the NF-kappa B signaling pathway. Specifically, HOXA10 up-regulated IKK beta gene transcription and sustained NF-kappa B activation in PDAC cells. Our findings suggest that HOXA10 functions as an oncoprotein in PDAC and may serve as a potential prognosis biomarker.

Systematic Identification of the RNA-Binding Protein STAU2 as a Key Regulator of Pancreatic Adenocarcinoma

Xiao Wang et al.

Summary: Pancreatic adenocarcinoma (PAAD) is a highly aggressive cancer. RNA-binding protein STAU2 plays a critical role in the occurrence and development of PAAD, and it can be a potential biomarker and therapeutic target for PAAD. High expression of STAU2 is associated with poor clinical outcomes and immune evasion in PAAD. Additionally, STAU2 is related to chemotherapy drug sensitivity, suggesting it could be used in combination therapy. This study provides valuable insights into the role of STAU2 in PAAD aggression and resistance, and it sheds light on its potential for cancer therapy and drug development.

Multiple Components of Protein Homeostasis Pathway Can Be Targeted to Produce Drug Synergies with VCP Inhibitors in Ovarian Cancer

Prabhakar Bastola et al.

Summary: The increased dependency of cancer cells on protein homeostasis creates a vulnerability that can be exploited using protein homeostasis inhibiting drugs. Recently, valosin-containing protein has emerged as a key component of the pathway and a novel therapeutic target for cancer treatment. Our results indicate that VCP inhibitors can be combined with other endoplasmic reticulum stress-inducing agents to produce synergistic cytotoxicity in ovarian cancer cells.
