Non-invasive prenatal screening for foetal trisomy: An assessment of reliability and reporting

Summary: This study evaluated the reliability and reporting standardization of non-invasive prenatal screening (NIPS) through an external quality assessment (EQA) program. Participating laboratories provided reliable results for samples with higher fetal fraction values, but false-negative results may be attributed to lack of performance validation, laboratory errors, and low fetal fraction values. In addition, enrichment operations should be validated and normalized, and further efforts are needed to standardize NIPS reports.

Cell-free DNA Testing in Routine Practice: Characterisation of a Cohort with Positive Results for Trisomies, Sex Chromosome Anomalies and Microdeletions

Summary: This study analyzed data from pregnant women with positive cfDNA results between September 2013 and December 2019, showing a PPV of 95% for T21 but lower rates for T18 and T13. Despite high specificity, the PPV for aneuploidies other than T21 is low in routine practice, and many patients did not receive a fetal anomaly scan before cfDNA testing.

Adverse pregnancy history is an important indicator for prenatal diagnosis, and nuchal fold thickening is closely related to fetal abnormalities. Utilizing CNV-seq in combination with ultrasound can improve the diagnosis of submicroscopic chromosomal aberrations in fetuses with congenital anomalies.
