HIV DNA positively correlates with HLA-DR+CD8+T lymphocytes over 8-year suppressive antiretroviral therapy

Summary: We conducted a study on the relationship between HIV DNA and CD8(+) T-cell activation in a retrospective cohort. Our findings showed that HLA-DR expression remained high in CD8(+) T-cells and was positively correlated with total HIV DNA (odds ratio = 1.86, P = 0.028) during 8 years of suppressive antiretroviral therapy (ART). This study highlighted the long-term interaction between HIV DNA reservoir and host inflammatory state despite viral suppression.

Summary: This study suggests that nonreplicating "defective" HIV-1 proviruses, which are transcriptionally and translationally competent, may contribute to persistent immune activation in people with HIV-1 on effective antiretroviral therapy. The presence of these proviruses and their correlation with antibody responses against HIV-1 proteins indicate a novel interplay between transcription and translation of defective proviruses and immune activation.
