Summary: The XCF3 groups (X = O, S, Se) play an increasingly important role in modern organic chemistry due to their unique electronegativity, lipophilic nature, metabolic stability, and bioavailability. The incorporation of XCF3 groups into heterocyclic compounds can change their physicochemical and biological properties, injecting new vitality into the application of heterocyclic compounds in various fields. This paper reviews the recent progress in the synthesis of F3CX-containing heterocycles and discusses the application scope and mechanism of some reactions.

Synthesis of medium-sized benzo[b]azocines and benzo[b]azonines by photoinduced 8-/9-endo sulfonyl-cyclization

Summary: An efficient photochemical radical sulfonyl cyclization method was developed for the synthesis of medium-sized benzo[b]azocines and benzo[b] azonines. This chemoselective method allows the synthesis of highly functionalized eight- and nine-membered N-heterocycles. Experimental studies and computational calculations were used to understand the radical mechanism and rationalize the observed chemoselectivity. The large-scale synthesis and derivatizations demonstrated the potential utility of this strategy for the synthesis of valuable medium-sized N-heterocycles.

The protocol involves an electrophilic cyclization process and is conducted under mild conditions. Mechanistic discussions rationalize the observed regioselectivity in the transformation. Further studies on the transformation of seleno-benzo[b]azepines and large-scale experiments demonstrate promising utility of this methodology.

Regioselective C6-H Hydroxyalkylation of Purines and Purine Nucleosides via a-C-H Functionalization of Alcohols at Room Temperature

Summary: In this study, a highly regioselective C-6-H hydroxylalkylation method for purines and purine nucleosides was reported for the first time, which was achieved via the alpha-C(sp(3))-H functionalization of alcohols at room temperature within 10 minutes. The reaction displayed tolerance towards various functional groups and had the potential for further modification to yield valuable molecules. Compared to other methods, this strategy provided a direct approach to access 6-substituted alkylated purines and nucleosides with pharmaceutical bioactivities, without the need for metal catalysts, light, and protecting groups.

Radical annulation of a designed diene system: access to nitro-benzo[b]azepines

Summary: In this study, a novel O2N-triggered ordered addition 7-endo cyclization reaction is described, showing excellent chemo- and regioselectivity. With this strategy, structurally diverse nitro-benzo[b]azepines were successfully prepared, with a total of 28 examples. The large-scale operation and convenient N-Ts and N-Cbz deprotection demonstrate the promising utility of this methodology. Mechanistic studies suggest that the reaction proceeds through a radical pathway.
