CuBr-promoted domino Biginelli reaction for the diastereoselective synthesis of bridged polyheterocycles: Mechanism studies and in vitro anti-tumor activities

Benchmarking Cleavable Biotin Tags for Peptide-Centric Chemoproteomics

Summary: Click chemistry, specifically the copper-catalyzed azide-alkyne cycloaddition, has allowed the development of chemical probes for functional proteome analysis. In this study, five commercially available cleavable biotin tags were evaluated in three common chemoproteomic workflows, resulting in a comparative analysis of 15 methods. An acid-cleavable biotin tag with a dialkoxydiphenylsilane moiety showed the best performance in terms of enrichment efficiency, identification yield, and reproducibility. Photocleavable and reduction-cleavable biotin tags can also be useful in certain scenarios. Furthermore, the systematic comparison of LC-MS/MS characteristics of tag-modified peptides provides valuable information for the future development of cleavable biotin reagents.
