Liver fibrosis therapy based on biomimetic nanoparticles which deplete activated hepatic stellate cells

Shenglong Xia et al.

Summary: Liver fibrosis is a common liver disease with significant morbidity and mortality. However, there is still a lack of effective therapy for liver fibrosis. In this study, biomimetic all-trans retinoic acid (ATRA) loaded PLGA nanoparticles (NPs) were developed to deplete activated hepatic stellate cells (aHSCs) by inducing apoptosis and quiescence. The NPs were coated with HSC membranes expressing tumor necrosis factor-related apoptosis-inducing ligand (TRAIL), and ATRA was encapsulated in the core. The biomimetic TM-ATRA/NPs showed the ability to induce both apoptosis and quiescence in aHSCs, leading to improved fibrosis amelioration in mouse models.

Targeting ER protein TXNDC5 in hepatic stellate cell mitigates liver fibrosis by repressing non-canonical TGFβ signalling

Chen-Ting Hung et al.

Summary: The study identified TXNDC5 as a critical mediator of liver fibrosis, showing that its upregulation is associated with the activation of HSCs and the production of excessive extracellular matrix. Targeting TXNDC5 could potentially be a novel therapeutic strategy to ameliorate liver fibrosis.

Reactive Oxygen Species-Responsive Polypeptide Drug Delivery System Targeted Activated Hepatic Stellate Cells to Ameliorate Liver Fibrosis

Yumei Hao et al.

Summary: Hepatic fibrosis is a chronic liver disease that lacks effective pharmacotherapeutic treatments. Researchers have designed targeted and reactive oxygen species (ROS)-responsive micelles for the specific delivery of resveratrol (RES), a traditional Chinese medicine, to activated hepatic stellate cells (aHSCs) for the treatment of liver fibrosis. In vitro and in vivo experiments demonstrate that these micelles enhance the targeted delivery of RES to aHSCs, reduce collagen accumulation, and protect hepatocytes from damage.

Polysaccharide-based supramolecular drug delivery systems mediated via host-guest interactions of cucurbiturils

Yiyun Gao et al.

Summary: Natural polysaccharides and their derivatives have great potential in drug delivery, with reversible crosslinking by Cucurbit[n]uril showing stimulus responsiveness for constructing stimuli-responsive drug release systems. The host-guest interactions of CB[6] and CB[8] in polysaccharide-based supramolecular drug delivery systems allow for easy tuning of rheological properties and drug release profiles.

ROS-responsive cyclodextrin nanoplatform for combined photodynamic therapy and chemotherapy of cancer

Die Jia et al.

Summary: Cyclodextrin is widely used for constructing CD-based nanoplatforms due to its special spatial structure and good biological safety. The study designed a highly sensitive ROS-responsive polymer PCP encapsulating doxorubicin (DOX) and purpurin 18 (P18) for synergistic photodynamic and chemotherapy effects. The micelles showed enhanced anti-cancer activity through the release of drugs triggered by ROS in the tumor microenvironment.

Molecular and cellular mechanisms of liver fibrosis and its regression

Tatiana Kisseleva et al.

Summary: Chronic liver injury results in liver inflammation and fibrosis, which can be reversed by eliminating activated myofibroblasts and resorbing the fibrous scar. Understanding the molecular mechanisms underlying liver fibrosis and its reversibility can lead to the identification of new therapeutic targets for liver fibrosis treatment.
