One-Pot Synthesis of α-Ketoamides from α-Keto Acids and Amines Using Ynamides as Coupling Reagents

Summary: A one-pot strategy using ynamides as coupling reagents has been developed for the formation of alpha-keto amide bonds under extremely mild reaction conditions. Diversely structural alpha-ketoamides were obtained in up to 98% yield for 36 examples. This reaction offers practical coupling procedure, wide functional group tolerance, and extremely mild conditions, with potential applications in synthetic and medicinal chemistry.

Visible-Light-Mediated Late-Stage Sulfonylation of Anilines with Sulfonamides

Summary: A visible-light-mediated late-stage sulfonylation of anilines with sulfonamides under simple reaction conditions is demonstrated. Various primary or secondary sulfonamides, including several pharmaceuticals, were successfully incorporated through N-S bond activation and C-H bond sulfonylation. The synthetic utility of this strategy is highlighted by the construction of complex anilines bearing diverse bioactive groups.

Gold(I)-Catalyzed Intramolecular Bicyclization: Divergent Construction of Quinazolinone and Ampakine Analogues

Summary: A gold(I)-catalyzed cascade intramolecular cyclization of Ugi adducts has been developed for the divergent construction of quinazolinone and ampakine analogues in a rapid, efficient and step-economical manner. This method shows high yields, excellent functional-group tolerance, broad substrate scope and excellent chemo- and regioselectivity. The practicality of this strategy is further demonstrated by a scale-up reaction. Diverse oligopeptides are also found to be well tolerated, affording polycyclic products decorated with a peptide chain.

Metal-Free Syntheses of α-Ketothioamide and α-Ketoamide Derivatives from Sulfoxonium Ylides
