LDLR missense variants disturb structural conformation and LDLR activity in T-lymphocytes of Familial hypercholesterolemia patients

Summary: Familial hypercholesterolemia (FH) is caused by harmful mutations in the LDLR gene, resulting in significantly increased low-density lipoprotein (LDL) cholesterol and premature atherosclerotic cardiovascular disease. In this study, the functional effects of LDLR variants in Brazilian FH patients were assessed. It was found that LDLR null and defective variants reduce LDL binding and uptake in activated T-lymphocytes, while LDLR missense variants affect LDLR conformational stability and the dissociation of the LDLR-ApoB complex.

Patients with high TG and low PE levels have shorter survival and poor prognosis.

Pharmacometabolomics for the Study of Lipid-Lowering Therapies: Opportunities and Challenges

Summary: Lipid-lowering therapies are widely used to prevent ASCVD and related mortality. Omics technologies have been employed to study the mechanisms and effects of these drugs, aiming to improve treatment efficacy and safety. Pharmacometabolomics focuses on studying the effects of drugs on metabolic pathways and the variation of treatment response. This review summarizes significant metabolomic studies on lipid-lowering therapies, including statins, fibrates, and novel drugs and nutraceutical approaches. Integrating pharmacometabolomics data with other omics approaches can help understand the biological mechanisms of lipid-lowering drugs and improve treatment efficacy while reducing side effects.

Summary: The statins, also known as HMG-CoA reductase inhibitors, are widely used as first-line therapy for hypercholesterolemia. Apart from their lipid-lowering effect, statins have been suggested to have various pleiotropic effects, including anti-inflammatory, antioxidant, vascular endothelial function-improving, plaque-stabilizing, and platelet aggregation-inhibiting effects. They have established preventive effects on atherothrombotic stroke and potential neuroprotective effects in other cerebrovascular diseases. Additionally, statins have shown pro-apoptotic, growth-inhibitory, and pro-differentiation effects in certain malignancies.

Genetic Variant ABCC1 rs45511401 Is Associated with Increased Response to Statins in Patients with Familial Hypercholesterolemia

Summary: In this study, the deleterious variant ABCC1 rs45511401 was found to significantly enhance the LDL-c response to statin treatment in Brazilian FH patients. However, deleterious variants in PK genes were not associated with an increased risk of statin-related adverse events. This suggests that ABCC1 rs45511401 may be a promising candidate for personalized statin therapy.
