Why does obesity cause diabetes?

Samuel Klein et al.

Summary: The accumulation of excessive body fat can lead to type 2 diabetes, and the risk of type 2 diabetes increases with an increase in body mass index. The global increase in obesity has therefore resulted in a corresponding increase in the prevalence of type 2 diabetes. The mechanisms linking obesity and type 2 diabetes involve complex changes in b cell function, adipose tissue biology, and multi-organ insulin resistance, which can be improved or even normalized through weight loss.

Molecular Mechanisms of Diabetic Kidney Disease

Jorge Rico-Fontalvo et al.

Summary: The inflammatory component of diabetic kidney disease is highly studied, with genetic and epigenetic variants playing a fundamental role in its development. Cells of the innate immune system contribute greatly to the pathogenesis, with a lesser role from adaptive immune cells. The complement system, cytokines, and chemokines also play a role in the disease.

Cornus officinalis vinegar reduces body weight and attenuates hepatic steatosis in mouse model of nonalcoholic fatty liver disease

Li Cao et al.

Summary: This study identified the bioactive components of Cornus officinalis vinegar (COV) and assessed its effects on body weight and hepatic steatosis in a nonalcoholic fatty liver disease (NAFLD) mouse model. The results showed that COV significantly reduced body weight, abdominal fat weight, liver weight, and improved liver function. COV also reduced glucose, triglyceride, and low-density lipoprotein cholesterol levels, while increasing high-density lipoprotein cholesterol levels. Additionally, COV increased anti-oxidant activity and reduced inflammation in the liver tissue of NAFLD mice.

ATP-citrate lyase inhibitor improves ectopic lipid accumulation in the kidney in a db/db mouse model

Zishun Zhan et al.

Summary: We evaluated a novel treatment for obesity-related renal issues by using an ATP-citrate lyase (ACL) inhibitor to reduce ectopic lipid accumulation in the kidney and the resulting inflammation. The results showed that the ACL inhibitor BMS-303141 had the highest affinity for ACL and could decrease ACL expression in the kidneys of db/db mice, leading to a reduction in renal ectopic lipid accumulation. Moreover, reducing ectopic lipid accumulation also improved renal injuries, inflammation, and tubulointerstitial fibrosis.

The Contribution of Lipotoxicity to Diabetic Kidney Disease

Jeffrey R. Schelling

Summary: This review focuses on the role of lipotoxicity in the pathogenesis of diabetic kidney disease (DKD). It highlights the lipotoxicity pathways in the podocyte and proximal tubule cell, which are considered as the most critical sites in DKD. The discussion also includes potential therapeutic targets and current pharmacologic approaches to reduce renal lipotoxicity.

Morroniside attenuates apoptosis and pyroptosis of chondrocytes and ameliorates osteoarthritic development by inhibiting NF-κB signaling

Huan Yu et al.

Summary: The study demonstrated that morroniside can attenuate the progression of osteoarthritis in mice by promoting cartilage matrix synthesis, inhibiting chondrocyte pyroptosis, reducing osteophyte formation, and subchondral sclerosis.

E2F1 promotes proliferation and metastasis of clear cell renal cell carcinoma via activation of SREBP1-dependent fatty acid biosynthesis

Donglai Shen et al.

Summary: Studies have shown that E2F1 plays a key role not only in regulating the cell cycle, but also in inducing lipid accumulation and elevated levels of lipogenic enzymes in ccRCC cells by upregulating SREBP1, promoting tumor proliferation and metastasis. Knockdown of E2F1 or suppression of SREBP1 can attenuate these effects in ccRCC cells.
