Synthesis and Anticancer Evaluation of New Indole-Based Tyrphostin Derivatives and Their (p-Cymene)dichloridoruthenium(II) Complexes

Summary: New N-alkylindole-substituted 2-(pyrid-3-yl)-acrylonitriles and their (p-cymene)Ru(II) piano-stool complexes were synthesized and found to exhibit potent antiproliferative activity in various cancer models. Some of the derivatives showed lower IC50 values than clinically relevant multikinase inhibitors gefitinib and sorafenib, indicating their potential as novel therapeutic agents for cancer treatment. The investigation of drug mechanism in HCT-116 p53-knockout colon cancer cells revealed that the derivatives induced apoptotic caspase-3/7 activity, ROS formation, and anti-angiogenic properties.

Influence of Indole-N Substitution of Thiosemicarbazones in Cationic Ru(II)(η6-Benzene) Complexes on Their Anticancer Activity

The substitution of a benzyl group at the indoleN improves the cytotoxic profile of the complexes compared to the allyl group, and the para-substituted benzyl scaffolds show higher anticancer activity than the meta-substituted compounds.

Half-Sandwich Ruthenium Arene Complexes Bearing Clinically Approved Drugs as Ligands: The Importance of Metal-Drug Synergism in Metallodrug Design

Summary: A novel strategy in metallodrug discovery is incorporating clinically approved drugs into metal complexes as ligands. This approach has been used to re-purpose drugs, prepare organometallic complexes, overcome drug resistance, and design promising alternatives to current metal-based drugs. The combination of organoruthenium moiety and clinical drug in a single molecule has been shown to enhance pharmacological activity and reduce toxicity. This review summarizes recent reports on rationally designed ruthenium complexes containing FDA-approved drugs, focusing on coordination mode, kinetics, mechanism, and structure-activity relationship. It provides insights for future developments in ruthenium-based metallopharmaceuticals.

The choice of μ-vinyliminium ligand substituents is key to optimize the antiproliferative activity of related diiron complexes

Summary: Diiron vinyliminium complexes are promising organometallics with potential anticancer activity. The synthesis and evaluation of [Fe2Cp2(CO)(mu-CO){mu-eta(1):eta(3)-C(R-3)C(R-4)CN(R-1)(R-2)}]CF3SO3 (2a-c, 4a-d) complexes were performed, including their solubility in D2O, Log P-ow, stability in D2O/Me2SO-d(6) mixture, and antiproliferative activity against ovarian cancer cell lines A2780 and A2780cisR, as well as the nontumoral cell line Balb/3T3 clone A31. The cytotoxicity data of 50 vinyliminium complexes were correlated with the structural properties, showing a positive correlation between octanol-water partition coefficient and relative antiproliferative activity on ovarian cancer cell lines. However, the effects of different substituents provided guidelines for the development of novel, more effective compounds. Three additional complexes (4p-r) were designed, synthesized, and biologically investigated, revealing their ability to inhibit thioredoxin reductase enzyme and induce cancer cell production of reactive oxygen species.

Cyanide-alkene competition in a diiron complex and isolation of a multisite (cyano)alkylidene-alkene species

Compound 3, with a multidentate alkylidene-alkene ligand, was found to be less stable than its geometric isomer 4 by 13.4 kcal mol(-1), and complete conversion to 4 was achieved by refluxing a THE solution of 3 for 2 hours.
