Estrogen deficiency accelerates postmenopausal atherosclerosis by inducing endothelial cell ferroptosis through inhibiting NRF2/GPX4 pathway

Ying Lv et al.

Summary: Oxidative stress and lipid metabolism disorder caused by estrogen deficiency contribute to postmenopausal atherosclerosis. This study used ovariectomized female ApoE(-/-) mice fed with a high-fat diet to simulate postmenopausal atherosclerosis. The progression of atherosclerosis was accelerated in these mice, accompanied by increased indicators of ferroptosis. Estradiol and a ferroptosis inhibitor showed protective effects against atherosclerosis and inhibited lipid peroxidation and iron deposition in endothelial cells. Additionally, estradiol exhibited anti-ferroptosis effects by improving mitochondrial dysfunction and upregulating GPX4 expression. NRF2 inhibition attenuated the protective effects of estradiol against ferroptosis. This research highlights the role of endothelial cell ferroptosis in postmenopausal atherosclerosis and the protective effects of NRF2/GPX4 pathway activation by estradiol.

Hydrogen sulfide protects Sertoli cells against toxicant Acrolein-induced cell injury

Zhimin Mao et al.

Summary: This study investigated the reproductive toxicity of acrolein (ACR) and the protective effect of hydrogen sulfide (H2S) in Sertoli cells and germ cells. It was found that ACR caused cell injury and oxidative stress in Sertoli cells, which was prevented by antioxidant treatment. H2S was shown to protect Sertoli cells and germ cells from ACR-induced cell death.

Hydrogen sulfide as a potent scavenger of toxicant acrolein

Zhimin Mao et al.

Summary: The study found that hydrogen sulfide (H2S) can effectively detoxify acrolein (ACR) toxicity, and increasing H2S levels can enhance cell resistance to ACR, while inhibiting H2S makes cells more sensitive. In animal experiments, inhibiting endogenous H2S increased sensitivity to ACR toxicity. Further analysis showed that H2S directly reacted with ACR, promoting ACR clearance and preventing protein carbonylation initiated by ACR.

ESR1 inhibits ionizing radiation-induced ferroptosis in breast cancer cells via the NEDD4L/CD71 pathway

Lin Liu et al.

Summary: This study found that overexpression of ESR1 is associated with decreased overall survival in estrogen receptor-positive breast cancer. There is a regulatory relationship between ESR1 and NEDD4L/CD71, which plays an important role in IR-induced ferroptosis and may be a potential target for radiotherapy in breast cancer.

Polysaccharide of Atractylodes macrocephala Koidz Alleviates Cyclophosphamide-Induced Thymus Ferroptosis in Gosling

Xiangying Zhou et al.

Summary: This study aimed to explore the mechanism by which Atractylodes macrocephala Koidz (PAMK) alleviates cyclophosphamide (CTX)-induced ferroptosis in thymocytes. The results showed that PAMK had a significant alleviating effect on thymus damage and thymocyte ferroptosis in goslings. Therefore, PAMK can be used as a natural alternative to antibiotics as a feed additive for immunomodulatory effects on goslings.

Doxorubicin causes ferroptosis and cardiotoxicity by intercalating into mitochondrial DNA and disrupting Alas1-dependent heme synthesis

Ko Abe et al.

Summary: The clinical use of doxorubicin is limited due to its cardiotoxicity. This study found that doxorubicin accumulates in mitochondria and induces iron-dependent cell death called ferroptosis in cardiomyocytes. Disruption of heme synthesis and impaired iron utilization contribute to this process. Additionally, administration of 5-aminolevulinic acid can prevent doxorubicin-induced cardiotoxicity.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.
