Design of a Coumarin-Based Fluorescent Probe for Efficient In Vivo Imaging of Amyloid-? Plaques

Summary: This study describes the design, synthesis, and evaluation of coumarin-derived small molecule fluorophores for A beta imaging. A novel fluorescence probe XCYC-3, which includes an aromatic coumarin framework embedded into the pi bridge of a push-pull chromophore, was found to efficiently recognize aggregated A beta and have good blood-brain barrier permeability. In vitro staining and in vivo imaging studies demonstrated its efficiency in recognizing A beta plaques in the brain of AD transgenic mice. These findings suggest that XCYC-3 is a promising fluorescence imaging agent for A beta and may contribute to the development of potent NIR fluorescent probes for A beta diagnosis in the future.

HOPE-SIM, a cryo-structured illumination fluorescence microscopy system for accurately targeted cryo-electron tomography

Summary: A cryo-correlative light and electron microscopy (cryo-CLEM) system named HOPE-SIM has been developed to efficiently target cryo-FIB for cryo-ET studies by incorporating a 3D structured illumination fluorescence microscopy (SIM) system and an upgraded high-vacuum stage. With the 3D super resolution of cryo-SIM and cryo-CLEM software 3D-View, the correlation precision of the targeting region of interest can reach 110 nm, sufficient for subsequent cryo-lamella fabrication. The HOPE-SIM system has been successfully utilized to prepare cryo-lamellae targeting mitochondria, centrosomes of HeLa cells, and herpesvirus assembly compartments of infected BHK-21 cells, demonstrating its high potential for future in situ cryo-ET workflows.

Wild-type α-synuclein inherits the structure and exacerbated neuropathology of E46K mutant fibril strain by cross-seeding

High spatial-resolution imaging of label-free in vivo protein aggregates by VISTA

Summary: Amyloid aggregation, a pathological hallmark in neurodegenerative diseases, was studied using label-free high-resolution imaging and machine learning for specific segmentation of aggregate cores and peripheral filaments. This non-invasive imaging technology allows precise and multiplex high-resolution imaging of protein aggregates and their micro-environment, opening up new biomedical applications.
