Summary: In this study, a tannic acid-Fe3+ metal-phenolic network (MPN)-based strategy is developed to convert the cytolytic peptide melittin (Mel) from foe to friend by reducing its hemolytic activity. The strategy uses strong interactions between proteins/peptides and polyphenols to shield Mel's positive charges. Additionally, an immune adjuvant resiquimod (R848) is introduced to enhance immunostimulation. The nanodrug created through this strategy effectively induces immunogenic cell death, stimulates antitumor immunostimulation, and shows potential for cancer treatment.

Rigid Shell Decorated Nanodevice with Fe/H2O2 Supply and Glutathione Depletion Capabilities for Potentiated Ferroptosis and Synergized Immunotherapy

Summary: A rigid metal-polyphenol shell decorated nanodevice ssPPE(Lap)@Fe-TA is constructed to overcome the limitations of ferroptosis therapy. It efficiently internalizes into tumor cells and initiates ferroptosis by disintegrating in the acidic endosome/lysosome. By depleting glutathione and promoting H2O2 release, it inhibits tumor progression through Fe3+-mediated Fenton reaction. Moreover, ssPPE(Lap)@Fe-TA induces a strong antitumor immune response and synergizes with anti-PD-L1 antibody to suppress tumor growth and metastasis.

Engineered anti-cancer nanomedicine for synergistic ferroptosis-immunotherapy
