Summary: Researchers developed an oxazine-based activatable molecular assembly (PTO-Biotin Nps) that can trigger lysosomal dysfunction-mediated Fenton pathway to induce tumor ferroptosis with excellent spatiotemporal resolution using near-infrared light. The assembly selectively accumulates in lysosomes of tumor cells and exhibits enhanced photothermal activity in the acidic tumor microenvironment. Upon activation, PTO-Biotin Nps induce lysosomal dysfunction, cytosolic acidification, impaired autophagy, and enhanced cellular Fenton reactions to induce ferroptosis, leading to improved antitumor efficacy and reduced systemic side effects.

Summary: In this study, an ingenious molecular engineering strategy was developed to regulate the cellular uptake and retention of rhodamine dyes. Introduction of polar aminoethyl increased the brightness and reduced the cellular uptake of dyes, and this change could be reversed by amino acetylation. Furthermore, these modifications enhanced the uptake of dyes by cancer cells (16-fold) through active transport. By constructing activatable probes, the signal contrast (56-fold) between cancer and normal cells was further improved, and it was confirmed that the released fluorophore could remain in cancer cells for an extended time, enabling long-term and specific tumor imaging.

Summary: In this study, an effective strategy to overcome the interference of albumin in biosystems for fluorophores/probes is proposed. This strategy utilizes an albumin-insensitive fluorophore and its changeable pi-conjugation. An oxazine-based fluorogenic probe for aminopeptidase N (APN) is designed to demonstrate this strategy. The probe is locked in a non-conjugated, colorless, and non-fluorescent state by modifying the N atom in the oxazine fluorophore with alanine through a cleavable linker, thereby eliminating the spectroscopic response caused by non-specific interaction with albumin. APN selectively cleaves the alanine moiety, restoring the large pi-conjugation and strong fluorescence. The capability of the probe to eliminate albumin influence is demonstrated by imaging APN in different cell lines and quantitatively determining APN in human serum and mouse urine. This strategy may be valuable for developing more specific fluorogenic probes for other enzymes.

Summary: This study successfully developed a series of two-photon small-molecule fluorogenic probes capable of sensitively detecting general NTR activities in various biological samples. The optimized probe, X4, displayed the best performance in terms of sensitivity and selectivity. X4 also showed excellent two-photon excited fluorescence properties and was able to directly monitor endogenous NTR activities in live cells, growing zebrafish, and tumor-bearing mice.
