Catalytic site-specific and regioselective (3+2) transannulations between 1,2,3-thiadiazoles and allenoates

Summary: In this study, tetrasubstituted thiophenes were produced in a redox-neutral manner through site-specific and regio-selective transannulation of Allenoates with 1,2,3-thiadiazoles in the presence of catalytic Rh(i)/bisphosphine. The 1,2,3-thiadiazoles act as masked 1,3-dipoles with nucleophilic sulfur and electrophilic carbon termini.

Rhodium-Catalyzed Transannulation of 1,2,3-Thiazoles with Alkynes for the Direct Synthesis of Thiochromenones

Summary: A rhodium-catalyzed denitrogenative carbonylation of 1,2,3-thiadiazole with alkynes is developed to construct (benzo)thiochromenones. This [3+2+1] cycloaddition is compatible with both terminal and internal alkynes, resulting in a wide range of poly-substituted thiochromenones. Unsymmetrical alkyne substrates show good regioselectivity. Notably, this strategy allows the synthesis of both monocyclic and benzo-thiochromenones, representing a novel example of denitrogenative transannulation with CO insertion.

Summary: This study developed a direct catalytic alkynylation/dehydrative cyclization method on water for the synthesis of a broad range of fluorinated 1,8-naphthyridines from 2-amino-3-trifluoroacetyl-pyridines and terminal alkynes. A unique N-heterocyclic carbene (NHC) ligand system was successfully utilized for copper- or silver-mediated cyclization. Computational analysis of the reaction pathway supported the explanation of different experimental conversions and yields for copper and silver catalysts.

Summary: In this study, ligand-controlled rhodium(I)-catalyzed denitrogenative transformations of different types of 4-vinyl-1,2,3-thiadiazoles were demonstrated. With [Rh(COD)(2)]BF4, vinylic 1,2,3-thiadiazoles undergo intramolecular transannulation reaction to afford substituted furans. However, with [Rh(COD)DPPF]BF4, intermolecular transannulation reaction occurs, providing access to highly functionalized thiophenes.

Radical Redox Annulations: A General Light-Driven Method for the Synthesis of Saturated Heterocycles

Summary: We introduce a two-component annulation strategy that involves the combination of aryl alkenes and redox-active radical precursors bearing tethered nucleophiles, enabling the synthesis of a diverse collection of five- and six-membered saturated heterocycles. This strategy employs an Ir(III) photocatalyst and a Bronsted acid under visible-light irradiation. The synthesis involves reductive proton-coupled electron transfer, radical addition to alkene, oxidative radical-polar crossover, and cyclization of the tethered nucleophile. This approach allows the easy access to a wide range of heterocycles, including pyrrolidines, piperidines, tetrahydrofurans, morpholines, delta-valerolactones, and dioxanones. It is amenable to gram-scale preparation and complex fragment coupling.

Synthesis of New 4-Vinyl-1,2,3-Thiadiazoles

Summary: Simple and convenient methods were developed for the synthesis of a variety of new 1,2,3-thiadiazole derivatives, including ethyl esters, alcohols, and aldehydes. Furthermore, twenty-nine structurally diverse 4-vinyl-1,2,3-thiadiazoles were successfully synthesized via Knoevenagel condensation. Some of the new vinylic derivatives were structurally confirmed using X-ray diffraction analysis.

Summary: A highly diastereoselective and chemoselective (3+2) transannulation reaction between 1,2,3-thiadiazoles and cyanoepoxides has been discovered, allowing the preparation of cis- and trans-products with high yields and selectivity. Bisphosphines play a dual role as ligands and catalysts, controlling the diastereoselectivity by catalyzing the isomerization of cis-epoxides. The steric hindrance of the epoxy group and its electron-withdrawing ability are key factors in the chemoselectivity and reactivity of the reaction.

Summary: In-depth mechanistic understanding of the Rh(i)-catalyzed transannulation of 1,2,3-thiadiazoles with alkenes, alkynes, and nitriles was gained through Density Functional Theory (DFT) calculations. The key intermediate in this reaction is suggested to be a four-membered cyclometalated Rh(iii) complex due to the easy chelation of the S atom with Rh. The regioselectivities in the product formation are not only determined by the feasible migratory insertion pathway, but also by the feasibility of the subsequent reductive elimination.
