Chemoenzymatic labeling of DNA methylation patterns for single-molecule epigenetic mapping

Summary: In this study, a chemoenzymatic procedure was developed for high-throughput epigenetic profiling of single DNA molecules. The method enables labeling of non-methylated CpGs with high efficiency and long DNA molecules. Using this approach, researchers successfully identified the decline in global DNA methylation in a chronic lymphocytic leukemia patient and performed whole-genome methylation mapping of the model plant.

Hydroxymethylation-Specific Ligation-Mediated Single Quantum Dot-Based Nanosensors for Sensitive Detection of 5-Hydroxymethylcytosine in Cancer Cells

Summary: In this study, a ligation-mediated nanosensor was developed for sensitive quantification of 5hmC modification in cancer cells. The nanosensor showed excellent specificity and high amplification efficiency, and could detect as low as 0.001% 5hmC DNA in complex mixtures. It also had the ability to monitor cellular 5hmC level and distinguish cancer cells from normal cells.
