Comparison of Four Immobilization Methods for Different Transaminases

Summary: This study aims to compare the stability of soluble and immobilized transaminases from different species. Immobilization of transaminases enabled their reusability for at least 10 cycles and maintained full or high activity. Upscaled kinetic resolutions demonstrated high conversion, maintained enantioselectivity, and high product yields, showing their applicability.

Co-Immobilization of D-Amino Acid Oxidase, Catalase, and Transketolase for One-Pot, Two-Step Synthesis of L-Erythrulose

Summary: In this study, an immobilized enzyme cascade was developed in a basket-type reactor for the one-pot, two-step enzymatic synthesis of L-erythrulose from D-serine and glycolaldehyde. The immobilized enzymes showed high catalytic activity and stability, leading to significantly improved L-erythrulose concentrations compared to separate immobilization of enzymes. The results demonstrate the potential of this method for the synthesis of a wide range of alpha-hydroxyketones.

Efficient Site-Selective Immobilization of Aldehyde-Tagged Peptides and Proteins by Knoevenagel Ligation

Summary: The aldehyde tag is used for selective labeling of proteins and preparing antibody-drug conjugates, or immobilizing enzymes or antibodies. A new solid support, modified agarose with an aryl substituted pyrazolone, allows site-selective and efficient immobilization of aldehyde-containing molecules under physiological pH without the need for additives or catalysts. Compared to reductive amination, the modified agarose achieves higher loadings and activities in various buffers at different concentrations and temperatures.

The enzyme was immobilized on a methacrylate-based carrier and showed higher stability compared to the soluble form in organic solvents. The immobilized enzyme also retained high activity after multiple synthesis cycles.

Chiral Synthesis of 3-Amino-1-phenylbutane by a Multi-Enzymatic Cascade System

Summary: In this study, asymmetric synthesis of 3-amino-1-phenylbutane (3-APB) was achieved by coupling a transaminase with a pyruvate decarboxylase (PDC) using an in situ by-product removal strategy. The addition of PDC significantly improved the synthesis of 3-APB under optimal reaction conditions, with yields higher than 60% and nearly 90% selectivity achieved. High stereoselectivity for (S)-3-APB was obtained using both CviTA and VflTA.
