From Initiation to Maintenance: HIV-1 Gp120-induced Neuropathic Pain Exhibits Different Molecular Mechanisms in the Mouse Spinal Cord Via Bioinformatics Analysis Based on RNA Sequencing

Jian Huang et al.

Summary: Human immunodeficiency virus (HIV) is a major health and development challenge that leads to neuropathic pain (HNP). Research has shown that the initiation and maintenance of HNP are regulated by different molecular mechanisms, providing potential molecular targets for HNP therapy.

Expression of Human Immunodeficiency Virus Transactivator of Transcription (HIV-Tat1-86) Protein Alters Nociceptive Processing that is Sensitive to Anti-Oxidant and Anti-Inflammatory Interventions

Thomas J. Cirino et al.

Summary: Despite successful cART in reducing viral load, a significant number of HIV+ patients report chronic pain. The exact mechanism of this comorbidity remains unknown, but the Tat protein of HIV is believed to play a role. Animal studies have shown that exposure to Tat protein can lead to pain and neuropathological manifestations similar to chronic pain reported by HIV+ patients.

Persistent sensory changes and sex differences in transgenic mice conditionally expressing HIV-1 Tat regulatory protein br

Summary: This study aims to investigate the role of chronic HIV-1 Tat exposure in the development of neuropathy in mice. The results show that Tat induces mechanical and cold hypersensitivity, with females exhibiting more severe hypersensitivity. Current therapeutics have limited effectiveness in alleviating symptoms in female mice.

Antibody Recognition of CD4-Induced Open HIV-1 Env Trimers

Zhi Yang et al.

Summary: The RV144 HIV-1 clinical vaccination trial showed modest protection against viral infection. Antibody responses to the V1V2 region of HIV-1 Env gp120 were correlated inversely with the risk of infection, and data from three other clinical vaccine trials suggested a similar signal. In this study, V2i antibodies and CD4i antibody CG10 were characterized for their interactions with open Env trimers bound to CD4. The findings provide insights into the immune mechanisms during HIV-1 infection.

An analgesic peptide H-20 attenuates chronic pain via the PD-1 pathway with few adverse effects

Long Zhao et al.

Summary: The lack of effective and safe analgesics for chronic pain management has long been a health issue. Analgesic peptides, without opioid-related adverse effects, have recently shown promise for pain treatment. This study discovers that the peptide H-20 binds to PD-1, reduces nociceptive signals, and displays effective analgesia in preclinical pain models, making it a promising candidate drug for chronic pain treatment.

Structural and Mechanistic Evidence for Calcium Interacting Sites in the HIV Transmembrane Protein gp41 Involved in Membrane Fusion

Yoel A. Klug et al.

Summary: In this study, calcium interaction motifs (CIMs) in the gp41 CHR and NHR regions were discovered via NMR spectroscopy. The assembly of CHR-NHR SHB was found to be facilitated in Ca2+-containing media and impaired in CIM mutants. The clinically approved fusion inhibitor T20 exhibited reduced inhibitory efficiency when challenged with calcium, which could have implications for the development of better fusion inhibitors for HIV.

When Physical and Social Pain Coexist: Insights Into Opioid Therapy

Mark D. Sullivan et al.

Summary: The opioid epidemic in the US challenges us to reconsider the function of opioids and the nature of chronic pain, highlighting the interconnection between physical and social pain. Patients using opioids long term for chronic pain treatment may experience both physical and social pain, with these medications potentially leading to persistent negative effects on the brain. Recognizing the social aspects of chronic pain and opioid action can lead to improved treatment and usage of opioid medications.

The Form and Function of PIEZO2

Marcin Szczot et al.

Summary: Mechanosensation is crucial for detecting dynamic mechanical stimuli such as pressure, stretch, and shear stress, with a breakthrough in 2010 being the discovery of mechanically gated ion channels called PIEZOs. The past decade of research has provided valuable information about the functional roles and mechanisms of these molecules, particularly PIEZO2, leading to a better understanding of touch and other types of mechanosensation.
