Mitochondrial ROS control neuronal excitability and cell fate in frontotemporal dementia

Noemi Esteras et al.

Summary: The study found that overproduction of mitochondrial reactive oxygen species (ROS) in 10+16 neurons alters the trafficking of specific glutamate receptor subunits, leading to impaired glutamatergic signaling, calcium overload, and excitotoxicity. Mitochondrial antioxidants can restore the altered response and prevent neuronal death. Additionally, extracellular 4R tau induces the same pathological response in healthy neurons, proposing a mechanism for disease propagation.

A Lipidomic Approach to Identify Potential Biomarkers in Exosomes From Melanoma Cells With Different Metastatic Potential

Simona Lobasso et al.

Summary: The study investigated the metastatic propensity of melanoma cells to bone colonization by analyzing the lipid profiles, revealing potential lipid biomarkers for different migration and invasiveness. A specific phospholipid was identified as a lipid marker of exosomes, showing differences in fatty acid tails and lipid composition between poorly metastatic and highly metastatic cells. The lipid profiles of exosomes derived from the two types of cells were not significantly different.

Lipid composition dictates the rate of lipid peroxidation in artificial lipid droplets

Mike Lange et al.

Summary: This study demonstrates that the lipid composition of lipid droplets (LDs) determines their sensitivity to lipid peroxidation (LPO). Increasing unsaturation levels in the phospholipid monolayer or triglyceride core of LDs increases their oxidation sensitivity, while the presence of cholesteryl esters has a dual effect depending on the acylated fatty acid. The ratio between different lipid species in LDs also plays a significant role in LPO propagation.

Summary: The study confirms the asymmetric distribution of cholesterol in the mammalian cell membrane, with significantly lower levels in the inner leaflet compared to the outer leaflet. Additionally, primary cells were found to have particularly low levels of cholesterol in the inner leaflet. These findings support the universality of low basal cholesterol concentration in the inner leaflet under physiological conditions.
