The mitochondrial Hsp70 controls the assembly of the F1FO-ATP synthase

Jiyao Song et al.

Summary: Mitochondrial Hsp70 plays dual roles in the formation of ATP synthase. It cooperates with Atp11 and Atp12 to form the F-1 domain, and it transfers Atp5 to link the catalytic head with the peripheral stalk. Inactivation of mtHsp70 leads to integration of assembly-defective Atp5 variants into the mature complex, indicating its quality control function.

β-Hydroxybutyrate impairs neutrophil migration distance through activation of a protein kinase C and myosin light chain 2 signaling pathway in ketotic cows

Yuxiang Song et al.

Summary: High concentrations of blood BHB impair PMN migration distance through inhibition of the trailing edge contraction, mediated by enhancing the activation of PKC-MLC2 signaling, which explains the dysfunctional immune state in ketotic cows and provides information on the pathogenesis of infectious diseases related to ketosis.

Impaired ketogenesis ties metabolism to T cell dysfunction in COVID-19

Fotios Karagiannis et al.

Summary: This study found that impaired production of beta-hydroxybutyrate (BHB) is associated with severe COVID-19 respiratory distress syndrome. BHB promotes the survival and interferon-gamma production of CD4(+) T cells. BHB acts as an alternative carbon source, fueling oxidative phosphorylation and maintaining the redox balance.

beta-hydroxybutyrate impairs monocyte function via the ROS-NLR family pyrin domain-containing three inflammasome (NLRP3) pathway in ketotic cows

Zhihao Dong et al.

Summary: This study investigated alterations in immune function, ROS production, and NLRP3 pathway activity in monocytes of cows with clinical ketosis. The results suggested that cows with ketosis had compromised immune functions with decreased phagocytosis ability and increased apoptosis and ROS content in monocytes.

Adaptation and Evolution of Pathogens in the Cystic Fibrosis Lung

Paul J. Planet

Summary: In contrast to acute respiratory infections, persistent bacterial infections in cystic fibrosis (CF) allow bacteria to evolve and adapt over time. This adaptation is reflected in mutations that accumulate in the genomes of the infecting bacteria. These mutations can lead to phenotypic differences such as antibiotic resistance or changes in the mucoid phenotype of Pseudomonas aeruginosa. Many of these adaptations occur repeatedly in different patients and even across different bacterial species.

The role of itaconate in host defense and inflammation

Christian G. Peace et al.

Summary: Inflammatory stimuli cause metabolic reprogramming in macrophages, particularly in the Krebs cycle, resulting in the accumulation of itaconate and other metabolites. Itaconate plays diverse roles in immunity and inflammation, including inhibiting inflammasome activation and glycolysis, activating anti-inflammatory transcription factors, and exerting anti-inflammatory effects in various disease models.

Itaconate confers tolerance to late NLRP3 inflammasome activation

Monika Bambouskova et al.

Summary: Itaconate, induced upon TLR stimulation, regulates NLRP3 inflammasome activation and synergizes with iNOS, establishing tolerance to NLRP3 inflammasome in inflammatory macrophages. Mechanistically, itaconate prevents full caspase-1 activation and gasdermin D processing, potentially leading to pyroptotic cell death and tissue damage if not controlled.

The metabolism-modulating activity of IL-17 signaling in health and disease

Rami Bechara et al.

Summary: IL-17, a cytokine discovered almost 30 years ago, has been found to play a crucial role in coordinating cellular and organismal metabolism. Understanding the relationship between IL-17 and downstream metabolic processes could potentially lead to new therapeutic opportunities for diseases associated with IL-17, even those not traditionally linked to this cytokine pathway.
