Unanswered questions in the regulation and function of the duplicated α7 nicotinic receptor gene CHRFAM7A

Sherry Leonard et al.

Summary: The alpha 7 nicotinic receptor is an important entry point for Ca2+ into the cell and is genetically linked to various diseases. The function of a recently discovered partial duplication CHRNA7, CHRFAM7A, negatively regulates the alpha 7 receptor and is associated with neurological and inflammatory disorders. More research is needed to understand its functional role, regulation, and its contribution to disease development. The review highlights unanswered questions and discusses a potential therapeutic approach using genome editing.

The Human-Restricted Isoform of the α7 nAChR, CHRFAM7A: A Double-Edged Sword in Neurological and Inflammatory Disorders

Simona Di Lascio et al.

Summary: CHRFAM7A is a human gene that arises from the partial duplication of the CHRNA7 gene, which is related to various disorders involving cognitive deficits. It plays an important role in extra-neuronal tissues, potentially modulating inflammatory responses and tumor progression. Studies suggest that the duplicated gene may negatively modulate alpha 7 activity and play a significant role in the therapeutic response to CHRNA7-related diseases.

Alpha-7 Nicotinic Receptor Dampens Murine Osteoblastic Response to Inflammation and Age-Related Osteoarthritis

Alice Courties et al.

Summary: This study investigated the role of non-neuronal cholinergic system in osteoarthritis (OA). The results showed that the cholinergic system, through alpha 7nAchR, can counteract the effects of inflammation on osteoblasts. Deletion of alpha 7nAchR may limit cartilage degradation in age-related OA. Therefore, enhancing the cholinergic system could be a potential therapeutic target, although it may depend on CHRFAM7A expression.

The emergence of the uniquely human α7 nicotinic acetylcholine receptor gene and its roles in inflammation

Wanling Peng et al.

Summary: The uniquely human CHRFAM7A gene, evolved from the fusion of two partially duplicated genes, encodes a duplicate alpha 7-nAChR that inhibits the channel functions of alpha 7-nAChR. The possible contribution of CHRFAM7A gene in disease research and drug discovery has not been fully considered.

CHRFAM7A Overexpression Attenuates Cerebral Ischemia-Reperfusion Injury via Inhibiting Microglia Pyroptosis Mediated by the NLRP3/Caspase-1 pathway

Xiangyuan Cao et al.

Summary: The study found that overexpression of CHRFAM7A attenuated cerebral ischemia-reperfusion injury by inhibiting microglia pyroptosis through the NLRP3/Caspase-1 pathway and promoting microglia polarization to the M2 phenotype.

Effect of CHRFAM7A Δ2bp gene variant on secondary inflammation after spinal cord injury

Mingkuan Lin et al.

Summary: The study revealed that in severe spinal cord injury patients, carriers of the Delta 2bp variant showed higher levels of circulating inflammatory mediators. On the other hand, in mild SCI patients, Delta 2bp carriers had lower levels of circulating IL-15. The temporal variation of inflammatory mediators post SCI was not significantly associated with the Delta 2bp variant.

Summary: The expression of cholinergic system members may be associated with COVID-19 severity and inflammatory response. The decreased expression of a negative dominant duplicate CHRFAM7A is correlated with COVID-19-induced hypercytokinemia.
