Summary: Ischemia/reperfusion (I/R) injury is a common cause of many diseases, leading to tissue damage and cell death. In this study, a cellular model was used to simulate retinal I/R injury and confirm the occurrence of pyroptosis as a regulated cell death mechanism. Through extensive analysis and experiments, it was found that DTX3L ubiquitinates NLRP3 to regulate pyroptosis induced by OGD/R injury in R28 cells.

Panaxadiol saponin ameliorates ferroptosis in iron-overload aplastic anemia mice and Meg-01 cells by activating Nrf2/HO-1 and PI3K/AKT/mTOR signaling pathway

Summary: Panaxadiol saponin (PND) is a potential targeted drug for the treatment of aplastic anemia (AA) that can suppress iron-induced ferroptosis in AA cells. PND improves iron deposition, labile iron pool, ferroptosis events, apoptosis, mitochondrial structure, and related markers in iron-induced cells. In an iron-overload AA mice model, PND ameliorates body weight, blood cell counts, BMMNCs, histological injury, and T lymphocyte percentage. Overall, PND attenuates ferroptosis in iron-overload AA cells and mice via activating Nrf2/HO-1 and PI3K/AKT/mTOR pathway, making it a promising therapeutic candidate for AA.
