m7G-quant-seq: Quantitative Detection of RNA Internal N7-Methylguanosine

Li-Sheng Zhang et al.

Summary: m7G-quant-seq is a quantitative method for accurately detecting and quantifying internal m7G sites in human cytoplasmic tRNAs. The method converts m7G sites into RNA abasic sites through efficient chemical reduction and mild depurination, and calculates the m7G modification fraction by analyzing the variation patterns during reverse transcription. The study demonstrates that m7G-quant-seq can successfully detect internal m7G sites in human cytoplasmic tRNAs and estimate the corresponding m7G methylation stoichiometry. This method is of great importance for monitoring the change in tRNA m7G methylation levels.

RNA partitioning into stress granules is based on the summation of multiple interactions

Tyler Matheny et al.

Summary: The study reveals that specific RNA and protein can influence the enrichment of RNA in stress granules. Artificial tethering of certain proteins can target mRNA into stress granules with multiple interactions required for efficient partitioning. This suggests a summation of RNA-protein, protein-protein, and RNA-RNA interactions collectively determine RNA recruitment into stress granules.

QKI is a critical pre-mRNA alternative splicing regulator of cardiac myofibrillogenesis and contractile function

Xinyun Chen et al.

Summary: The RNA-binding protein Quaking (QKI) plays a crucial role in cardiac development and contractile physiology through its regulation of alternative splicing in genes involved in Z-disc formation and contractile physiology. Deficiency of QKI results in the failure of cardiomyocytes to mature into functional cells, highlighting its importance in cardiac pathophysiology.

N7-Methylguanosine tRNA modification enhances oncogenic mRNA translation and promotes intrahepatic cholangiocarcinoma progression

Zihao Dai et al.

Summary: Intrahepatic cholangiocarcinoma (ICC) shows upregulation of N-7-methylguanosine (m(7)G) tRNA modification and its methyltransferase complex components, METTL1 and WDR4, which are associated with poor prognosis. METTL/WDR4 play a critical role in promoting ICC cell survival and progression by selectively regulating the translation of oncogenic transcripts through m(7)G-tRNA-decoded codon-frequency-dependent mechanisms. Studies with overexpression and knockout mouse models demonstrate the crucial oncogenic function of Mettl1-mediated m(7)G tRNA modification in promoting ICC tumorigenesis and progression in vivo.
