Concurrent outcomes from multiple approaches of epistasis analysis for human body mass index associated loci provide insights into obesity biology

Sheldon D'Silva et al.

Summary: Genome wide association studies (GWAS) focus on single variant effects in additive genetic models, but only explain part of the heritability of complex traits. This study conducted epistasis analysis for BMI associated SNPs and identified two significant pairwise interactions, highlighting the influence of neuronal genes and implicating the coordinated expression of associated genes in liver, pancreas, and adipose tissues.

The sequences of 150,119 genomes in the UK Biobank

Bjarni Halldorsson et al.

Summary: A comprehensive understanding of how diversity in the human genome sequence affects phenotypic diversity relies on a reliable characterization of both sequences and phenotypic variation. In this study, whole-genome sequencing of 150,119 individuals from the UK Biobank was performed, leading to insights into the relationship between sequence variation and phenotypic traits. The analysis revealed rare variants with large effects, which were not previously identified through whole-exome sequencing and/or imputation studies.

Variance-quantitative trait loci enable systematic discovery of gene-environment interactions for cardiometabolic serum biomarkers

Kenneth E. Westerman et al.

Summary: Gene-environment interactions play a critical role in understanding diseases and personalized medicine. In this study, researchers performed a genome-wide analysis and identified several loci-biomarker pairs that interact with environmental factors. This catalog of gene-environment interactions provides valuable insights into the impact of genetics and environment on health.

Multiomics study of nonalcoholic fatty liver disease

Gardar Sveinbjornsson et al.

Summary: This study identified genetic variants, genes, and proteins associated with nonalcoholic fatty liver, cirrhosis, and hepatocellular carcinoma. The findings provide insights into the development of noninvasive evaluation and new therapeutic options for NAFL. Proteomics can also distinguish between NAFL and cirrhosis.

The power of genetic diversity in genome-wide association studies of lipids

Sarah E. Graham et al.

Summary: This study conducted a multi-ancestry, genome-wide genetic discovery meta-analysis of lipid levels in approximately 1.65 million individuals, including 350,000 of non-European ancestries. The findings demonstrate that increasing diversity rather than studying additional individuals of European ancestry results in substantial improvements in fine-mapping functional variants and portability of polygenic prediction, with modest gains in the number of discovered loci and ancestry-specific variants. This highlights the importance of including diverse populations in genetic studies to improve the accuracy and equity of polygenic scores in clinical practice.

Pairwise effects between lipid GWAS genes modulate lipid plasma levels and cellular uptake

Magdalena Zimon et al.

Summary: Studying the contribution of pairs of genes to complex traits has been challenging. Here, the authors combine exome and genotype data with RNAi to screen for genetic interactions between 30 genes identified in lipid GWAS to hint at pairs whose joint modulation may improve lipid-lowering therapies.

Genome-wide association study of fish oil supplementation on lipid traits in 81,246 individuals reveals new gene-diet interaction loci

Michael Francis et al.

Summary: Fish oil supplementation has mixed effects on circulating cardiovascular biomarkers, with genetic polymorphisms playing a role in the response of blood lipids to supplementation. This study identified four genetic loci where fish oil supplementation can modify the effects on blood lipids, highlighting the potential for personalized nutrition interventions to reduce cardiovascular disease risk.
