The KMT2A recombinome of acute leukemias in 2023

C. Meyer et al.

Summary: This study analyzed the genomic breakpoints in 3401 acute leukemia patients and identified 107 KMT2A gene fusions and other rearrangements. The study also found seven common fusion genes and partial tandem duplications, which are important for understanding the pathogenesis of acute leukemia and monitoring minimal residual disease.

Plasticity of Extrachromosomal and Intrachromosomal BRAF Amplifications in Overcoming Targeted Therapy Dosage Challenges

Kai Song et al.

Summary: Understanding the structure and dynamics of oncogene amplifications is crucial for overcoming tumor relapse. BRAF amplifications in melanoma show high plasticity under MAPKi dosage challenges, involving de novo genomic alterations even in the HSR mode. Furthermore, BRAF FA-driven, dual MAPKi-resistant cells expand the spectrum of resistance-linked ferroptosis sensitivity.

Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Dynamic regulation of CTCF stability and sub-nuclear localization in response to stress

Bettina J. Lehman et al.

Summary: The study reveals that CTCF is highly sensitive to diverse forms of stress in human primary cells, leading to changes in its localization and interactions with RNA. The stress-sensitive pool of CTCF protein can be restored through proteasome inhibition. This stress response in CTCF is observed during neuronal differentiation, highlighting its potential role in cell differentiation regulation.

Collective regulation of chromatin modifications predicts replication timing during cell cycle

Capucine Van Rechem et al.

Summary: The study investigates the relationship between replication timing and genome architecture, finding four primary chromatin states and an uncharacterized H3K36me2 state. It also reveals that changes in broad histone marks strongly impact replication timing.

Assessing acute myeloid leukemia susceptibility in rearrangement-driven patients by DNA breakage at topoisomerase II and CCCTC-binding factor/cohesin binding sites

Naomi D. Atkin et al.

Summary: This study developed a highly sensitive method for mapping DNA breaks in patient samples and found significantly increased DNA breakage at the KMT2A gene region in fusion-driven AML patients. Additionally, specific DNA break-prone regions were identified, providing greater predictive power for evaluating individual susceptibility.
