Coordination Chemistry of Nucleotides and Antivirally Active Acyclic Nucleoside Phosphonates, including Mechanistic Considerations

Astrid Sigel et al.

Summary: The coordination chemistry of nucleotides and their derivatives is crucial in the field of biological inorganic chemistry, as nucleotides play a direct or indirect role in all natural processes. Chemical modifications of nucleotides have led to the discovery of medically useful compounds, such as acyclic nucleoside phosphonates (ANPs), which exhibit antiviral activity. Among ANPs, diphosphorylated PMEA has been found to be a better substrate for polymerases compared to ATP due to its conformation, increased metal ion affinity, and the formation of a 5-membered chelate. Understanding the coordination chemistry of nucleotides and their derivatives can contribute to the development of new drugs and therapies.

Design and Synthesis of New α-hydroxy β-fluoro/β-trifluoromethyl and Unsaturated Phosphonates from Carbohydrate-Derived Building Blocks via Pudovik and Horner-Wadsworth-Emmons Reactions

Monika Bilska-Markowska et al.

Summary: In this study, we applied fluorinated carbohydrate-derived building blocks for the synthesis of alpha-hydroxy beta-fluoro/beta-trifluoromethyl and unsaturated phosphonates. Pudovik and Horner-Wadsworth-Emmons reactions were used to achieve this goal. The proposed pathway of the key reactions was supported by experimental results and quantum chemical calculations. The structures of the products were determined by spectroscopic and spectrometric techniques. Based on our findings, we conclude that the progress of the Pudovik and HWE reactions is influenced by acidic protons present in the molecules.

Highly Z-Selective Horner-Wadsworth-Emmons Olefination Using Modified Still-Gennari-Type Reagents

Ignacy Janicki et al.

Summary: This report presents new, easily accessible reagents for highly Z-selective HWE reactions, demonstrating very good Z-selectivity and high yields in most cases. The application of these new reagents may provide a valuable and practical alternative to the well-established Still-Gennari or Ando Z-selective carbonyl group olefination protocols.

Applications of the Horner-Wadsworth-Emmons Olefination in Modern Natural Product Synthesis

David Roman et al.

Summary: The Horner-Wadsworth-Emmons (HWE) reaction is a reliable olefination reaction widely used in organic chemistry and natural product synthesis, with excellent selectivity. This review summarizes the application of HWE olefinations in total syntheses of structurally different natural products, serving as a guideline for future synthetic approaches.
